摘要
目的研究高良姜素促进人乳腺癌细胞株MCF-7的凋亡作用。方法采用CCK-8法检测高良姜素对MCF-7细胞的增殖抑制作用;以流式细胞术检测细胞凋亡;Western blot法检测Bax、Bcl-2、cleavedcaspase-3和cleaved-PARP等相关蛋白的表达水平。结果高良姜素对MCF-7细胞增殖有明显的抑制作用,24 h和48 h的IC50分别为43.71μmol·L^(-1)和20.68μmol·L^(-1);高良姜素能够诱导MCF-7细胞凋亡,呈浓度依赖关系;高良姜素能上调Bax、cleaved-caspase-3和cleaved-PARP蛋白的表达,下调Bcl-2蛋白的表达。结论高良姜素对MCF-7细胞增殖有明显的抑制作用,通过线粒体途径调节凋亡相关蛋白的表达水平诱导MCF-7细胞凋亡。
Objective To explore the effect of galangin on inducing the apoptosis of human breast cancer cells MCF-7.Methods The effect of galangin on inhibiting MCF-7 proliferation was determined by CCK 8. The cell apoptosis was detected by flow cytometry with FITC-Annexin V/PI. The expression levels of Bax,Bcl-2,cleaved-caspase-3 and cleaved-PARP protein were detected by Western blotting. Results Galangin could obviously inhibited the proliferation of MCF-7,and the IC50 at 24 h was 43.71 μmol·L(-1) and that at 48 h was 20.68 μmol·L(-1). Galangin induced the apoptosis of MCF-7 cells in dose-dependent manner, and up-regulated the expression levels of Bax, cleavedcaspase-3 and cleaved-PARP protein while down-regulated the expression of Bcl-2. Conclusion Galangin shows obvious inhibitory effect on the proliferation of MCF-7, and can induce the apoptosis of MCF-7 by regulating apoptosis-associated proteins through mitochondrial pathway.
