基于黑果枸杞药材指标性成分的含量测定研究 被引量:5

摘要 目的:建立采用HPLC-ELSD法和苯酚-硫酸法分别测定13批黑果枸杞药材中甜菜碱和多糖含量的方法。方法:以Hypersil NH_2(250mm×4.2mm,5μm)柱为色谱柱,乙腈-水(83∶17)为流动相,等度洗脱,流速为0.7mL·min^(-1),柱温为35℃,载气流速为2.0L·min^(-1),漂移管温度为80℃,增益值为1,对甜菜碱进行定量分析。采用水提醇沉法制备供试品溶液,以苯酚-硫酸显色,采用紫外分光光度法测定多糖含量。结果:甜菜碱在0.01~0.1mg·mL^(-1)范围内线性关系良好,平均回收率为100.5%,RSD为1.2%。葡萄糖对照品在20~100μg/mL范围内线性关系良好,平均回收率为100.50%,RSD为2.61%。结论:所建立的含量测定方法稳定可靠,可对黒果枸杞药材的质量进行分析与评价。 Objective:To establish a method of HPLC-ELSD and phenol sulfuric acid method for determination of betaine and polysaccharide in 13 batches of Lycium ruthenicum Murr.Methods:Hypersil NH_2(250nm*4.2mm,5μm)column,acetonitrile water(83:17)as mobile phase,gradient elution.The flow rate was 0.7mL·min^(-1),the column temperature is 35 DEG C,carrier gas flow rate is 2.0 L·min^(-1),the drift tube temperature is 80 DEG C,the gain value is 1,the betaine quantitatively analysis.Using water extraction and alcohol precipitation to prepare the sample solution,the content of polysaccharide was determined by UV spectrophotometry.Results:Betaine was good in the range of 0.01~0.1mg.mL^(-1),the average recovery rate was 100.5%,and the RSD was 1.2%.In the range of 20~100g/mL,the glucose control was good,the average recovery was 100.50% and RSD was 2.61%.Conclusion:The established HPLC method is stable and reliable,can analyze and evaluate the quality of Lycium ruthenicum Murr.medicinal herbs.
