摘要
目的:优选钩藤生物碱的最佳提取工艺。方法:以钩藤总碱的含量,钩藤碱、异钩藤碱、去氢钩藤碱和异去氢钩藤碱的含量总和及浸膏得率为评价指标,对比了水煮、渗漉、索式回流和回流四种方法的提取效果,优选出回流的提取方法,并采用正交设计的方法考察溶剂浓度、溶剂倍量、提取时间及提取次数对钩藤生物碱的提取率和含量的影响。结果:钩藤生物碱的最佳提取工艺为:10倍量90%乙醇,回流提取2次,每次1h。结论:优选确定的工艺简便、稳定可行。
Objective:To Investigate the best extraction process of alkaloid from Uncaria rhynchophyila. Methods:With The con tent of total alkaloids, rhynchophylline,isorhynchophylline, corynoxeine and isocorynoxeine, and dry extract rate as the evaluating in- dicators, compared the extraction effect of the decocting method, the percolation method, soxhlet extraction and the ethanol refluxing method. The ethanol refluxing method was the best way. The orthogonal test method was adopted to investigate the influence of four extracting factors (solvent concentration, solvent quantily, extraction time, extraction times) on the extraction rate and content of Uncaria alkaloids. Results:The optimum extraction process condion was:10 times amount of 90 % ethanol, extracting 2 times for 1 hours eaehextraction. Conclusion : This extract process is simple, stable and feasible.
