摘要
目的探讨调节性T细胞(regulatory T cell,Treg)在年轻女性宫颈鳞状上皮内病变(cervical squamous intraepithelial lesions,CSIL)、宫颈癌组织中的表达及其与不同病变级别的相关性,并研究其预测CSIL转归的作用。方法选取2014年6月-2016年6月在广东省妇幼保健院或深圳市妇幼保健院就诊的年龄<35岁CSIL、宫颈癌患者110例,分为宫颈低级别鳞状上皮内病变(Low-grade squamous intraepithelial lesion,LSIL)组(30例)、宫颈高级别鳞状上皮内病变(High-grade squamous intraepithelial lesion,HSIL)组(30例)及宫颈癌组(50例),另选取同期因其他疾病(如子宫肌瘤、子宫内膜异位症)行全子宫切除的正常宫颈组织30例为正常对照组。进行免疫组织化学染色,检测宫颈组织中叉头状旋转录因子3(forkhead helix transcription,FOXP3)的表达情况。结果 FOXP3在正常对照组、LSIL组、HSIL组及宫颈癌组中的阳性表达率分别为3.3%、36.6%、53.3%和100.0%,FOXP3在不同级别CSIL、宫颈癌中的总阳性率有显著差别,疾病级别越重,总阳性率越高。正常对照组、LSIL组、HSIL组及宫颈癌组病变组织中,FOXP3的阳性表达强度与病变级别相关(P<0.001)。结论 FOXP3在年轻女性CSIL、宫颈癌组织中阳性表达强度与病变级别相关,可辅助临床评价肿瘤的生物学行为。
Objective To explore the expressions of regulatory T cells in cervical squamous intraepithelial lesions(CSIL) and cervical carcinoma in young women and the correlations with different degrees of cervical lesions,research the predictive function for prognosis of CSIL.Methods A total of 110 cervical carcinoma patients less than 35 years old were selected from the hospital from June 2014 to June2016,then they were divided into low-grade squamous intraepithelial lesion(LSIL) group(30 patients),high-grade squamous intraepithelial lesion(HSIL) group(30 patients),and cervical carcinoma group(50 patients).Thirty patients with normal cervix after total hysterectomy due to other diseases(uterine fibroids,endometriosis) during the same period were selected as control group.Immunohistochemical staining was used to detect the expression of forkhead/winged helix transcription factor 3(FOXP3) in cervical tissues.Results The positive expression rates of FOXP3 in control group,LSIL group,HSIL group,and cervical carcinoma group were 3.3%,36.6%,53.3%,and 100.0%,respectively,there were significant difference,the severer the lesions were,the higher the total positive expression rate was.The positive expression intensity of FOXP3 was correlated with the grades of lesions in control group,LSIL group,HSIL group,and cervical carcinoma group(P〈0.001).Conclusion The positive expression intensity of FOXP3 was correlated with the grades of lesions in young women with CSIL and cervical carcinoma,which can help clinical evaluation of biological behaviors of cervical tumors.
