茄叶片中蛋白质晶体的超微观察

摘要 为了解茄叶片内蛋白质晶体的几何形状和切面的超微特征,采用透射电子显微技术对茄叶片中的蛋白质晶体进行了超微观察。结果表明,叶片不同类型的细胞中都含有蛋白质晶体,晶体只发生于过氧化物酶体中,占据过氧化物酶体绝大部分内部空间,晶体大小与过氧化物酶体大小相对应。晶体切面呈现规则的几何形状,包括梯形、平行四边形、长方形、正方形、五边形和六边形,这些形状为平行六面体不同角度切割的结果;晶体切面具有清晰的条纹,条纹相互平行。由此得出结论,茄叶片中蛋白质晶体呈平行六面体形,具有网格状超微特征,其发生没有组织特异性,但有细胞内部空间特异性,只发生于过氧化物酶体中。 Transmission electron microscopy was applied to characterize protein crystals in leaf blades of Solanum melongena, with the aim to understand whether the occurrence of protein crystals has tissue and intracellular compartment specificity and what shape the crystals take. It was demonstrated that protein crystals occurred in all types of cells. Protein crystals were present only in peroxisomes and occupied most of their volume, and no protein crystals were found in other intracellular compartments. Protein crystals had varied sizes, dependent upon the size of peroxisomes where they were located. The 2-D images of protein crystals were trapezoid, parallelogram, rectangle, square, pentagon, and parallelepiped. These 2-D images suggested a parallelepiped 3-D images of protein crystals. The sections of the crystals revealed a striped pattern. In conclu- sion, the occurrence of protein crystals in Solarium melongena leaf blades did not have tissue but intracellular compartment specificity, and the crystals took a parallelepiped shape and their sections had a striped uhrastrnctural pattern.
