石橄榄种子无菌播种和快速繁殖研究 被引量:3

摘要 [目的]建立石橄榄种子无菌播种和快速繁殖技术体系。[方法]以MS为基础培养基,添加不同浓度6-BA、NAA、有机添加物,筛选适宜的诱导、增殖及壮苗生根培养基组合。[结果]以MS+6-BA 1.0 mg/L+NAA 0.5 mg/L+椰子汁50 mL/L为石橄榄种子最佳诱导培养基,14 d可见种子萌动,45 d可分化形成丛生幼苗;以MS+6-BA 1.0 mg/L+NAA 0.1 mg/L+椰子汁50 mL/L为最佳增殖培养基,其增殖倍数可达2.750倍,假鳞茎形成率达24.5%;以1/2MS+NAA 0.1 mg/L+AC 1 g/L为最佳壮苗生根培养基,苗高可达3.82 cm,根数1.9条。[结论]该研究为石橄榄野生种质资源保护、快速繁育优质种苗奠定了基础。 [Objective] To establish a system of aseptic sowing and rapid progagation technology for Pholidota chinensis Lindl..[Method]Different concentrations 6-BA,NAA,organic additives were added to MS medium to screen the suitable induction,proliferation and strong seedling and rooting medium.[Result]The proper medium for the seeds induction of P.chinensis Lindl.was MS medium containing 6-BA 1.0 mg/L+NAA 0.5 mg/L+coconut juice 50 mL/L,seed germination was observed on 14 days,the differentiation and formation of clusters of seedlings were observed on 45 days;the best multiplication medium was MS medium containing 6-BA 1.0 mg/L+NAA 0.1 mg/L+coconut juice 50 mL/L,proliferation times 2.750,the pseudobulb formation rate up to 24.5%;the best seedling rooting medium was 1/2MS medium containing NAA 0.1 mg/L+AC 1 g/L,plant length up to 3.82 cm,root number 1.9.[Conclusion]The study laid a foundation for the conservation of wild germplasm resources and rapid breeding of high quality seedling.
