摘要
目的探讨丙酮酸乙酯对滑膜成纤维细胞增殖的影响。方法收集胶原诱导的关节炎大鼠(collagen induced arthritis,CIA)和正常大鼠滑膜组织,培养滑膜成纤维细胞。光镜及免疫荧光法检测Vimentin蛋白鉴定滑膜成纤维细胞;分别用1、2、5、10、20 mmol/L浓度的丙酮酸乙酯作用于滑膜成纤维细胞;流式细胞术及CCK8法检测细胞增殖情况;流式细胞术检测细胞凋亡情况。结果显微镜观察细胞形态特征符合滑膜成纤维细胞,且Vimentin蛋白表达阳性;5种剂量的丙酮酸乙酯均能抑制细胞增殖同时诱导细胞凋亡,但是未见明显剂量依赖效应,以5 mmol/L剂量组作用效果最为明显。结论丙酮酸乙酯能够抑制滑膜成纤维细胞增殖同时促进其凋亡,且以5 mmol/L剂量组作用最为明显。
This study was performed to explore the effect of ethyl pyruvate on the proliferation and apoptosis of synovial fibroblasts. The knee synovial tissues of CIA and normal rats were collected and the synovial fibroblasts were cultured. Then the synovial fibroblasts were identified with microscope and immunofluorescence. Ethyl pyruvate of 1 mmol/L, 2 mmol/L, 5 mmol/L, 10 mmol/L and 20 mmol/L were used to treat synovial fibroblasts, then the cell proliferation and apoptosis of the synovial fibroblasts were detected by flow cytometry/CCK8 assay and flow cytometry, respectively. Data showed that the morphology and characteristics of the cells were accorded with synovial fibroblasts, and most cells expressed vimentin. All five dosage of ethyl pyruvate could inhibit cell proliferation and induce the apoptosis of synovial fibroblasts, but not showing a dosage-dependent manner. 5 mmol/L-dosage of ethyl pyruvate could remarkably inhibit cell proliferation and induce cell apoptosis. This study suggest that ethyl pyruvate could inhibit cell proliferation and induce the apoptosis of synovial fibroblasts, and the effect is most obvious at 5 mmol/L-dosage group.
