诱导多潜能干细胞来源的神经干细胞在阿尔茨海默病模型及其治疗方面的研究进展

摘要 阿尔茨海默病(AD)是最常见的神经退化性疾病,临床表现主要为记忆力逐渐丧失和认知功能障碍。其药物治疗效果不佳,且临床试验取得的成果少,但利用神经干细胞(NSCs)移植到AD小鼠模型的脑部,可恢复部分行为学功能。由于诱导多潜能干细胞(iPSCs)来源的NSCs,具有分化效率高、不存在排斥反应及伦理问题等优点,因而为神经退行性疾病提供了新的治疗策略。故探讨iPSCs向NSCs分化及提高其分化效率的方法,对iPSCs及由iPSCs来源的NSCs分化的神经元细胞在AD模型以及治疗方面的研究具有重要意义。 Alzheimer's disease (AD) is the most common form of senile dementia, the clinical features of which mainlyinclude gradual memory loss and cognitive dysfunctionThe efficacy of drug therapy for AD is poor and the chnical trials haveseen few results, but with the transplantation of neural stem cells (NSCs) into the brain of AD mouse model, it is found thatsome behavioral functions can be restored. NSCs derived from induced pluripotent stem cells (iPSCs) are characterized byhigh differentiation efficiency, no immuno-rejection and ethical issues etc. , which provides and establishes the new opportu-nity for the treatment of neurodegenerative diseases. Therefore,it is important to explore the methods of iPSCs to NSCs differ-entiation and improve its differentiation efficiency, and the study of IPSCs and iPSCs-derived NSCs differentiation of neuroncells in AD model and treatment is of great significance.
