一株中度嗜盐菌新种LYG2#的分离与鉴定

摘要 [目的]对一株从连云港青口卤水中分离纯化出的细菌LYG2#进行分类学鉴定。[方法]通过16S rRNA基因序列比对、系统发育分析和一系列生理生化指标测定对该细菌LYG2#进行鉴定。[结果]该菌为革兰氏阴性菌,菌落形态为圆形,粉红色。电镜下观察菌体为螺旋状,长1.50~2.00μm,宽0.24μm。LYG2#最适生长盐浓度为9.6%,最适生长温度为32℃,最适pH为8.5。通过16S rRNA基因序列比对,发现其与Spiribacter salinus M19-40~T的相似度最高,为95.98%。菌株LYG2#在中国科学院的保藏号为CGMCC 1.12136,Gen Bank序列登录号为JQ087462。[结论]分离的菌株LYG2#为一株中度嗜盐菌,是Spiribacter属的一个新种资源。 [Objective] To identify a strain LYG2# isolated fromLianyungang Qingkou Salinas.[Method]We identified the strain LYG2# by means of 16 S rRNA gene sequence blast,phylogenetic analysis and the determination of physiological and biochemical characteristics.[Result] The bacteria was gram negative,appeared pink and round colony.Strain LYG2# was spiral,the length of the bacteria was from 1.50 to2.00 μm,and the width was 0.24 μm.The optimum salt concentration,temperature and pH were 9.6%,32℃ and 8.5.By 16 S rRNA gene sequence blast,the 16 S r DNA of the strain LYG2# and Spiribacter salinus M19-40 Tshowed the highest similarity,was 95.98%.Strain LYG2# was preserved at China General Microbiological Culture Collection Center,the number was CGMCC 1.12136,the Gen Bank accession number for 16 SrDNA of the strain was JQ087461.[Conclusion]The bacteria isolated was a moderately halophilic bacterium,and was a new species of the Spiribacter genus.
