响应面优化热浸提法提取石榴皮总多酚的工艺研究 被引量:7

摘要 [目的]研究石榴皮中总多酚的热浸提最佳工艺条件并测定其含量。[方法]以石榴皮为原料、纯水为浸提溶剂加热浸提石榴皮中总多酚,通过单因素试验探究了石榴皮总多酚提取率随热浸提温度、时间、料液比以及提取次数4个客观因素的变化规律,并用响应面分析法优化提取工艺,采用Folin-Ciocalteu比色法测定石榴皮总多酚含量。[结果]通过Box-Benhnken响应面分析试验确定以热浸提法提取石榴皮总多酚的最佳工艺为热浸提温度45℃、时间30 min、料液比1∶20(g∶mL)、提取次数为3次,4个因素对石榴皮总多酚提取率的影响顺序依次为温度、料液比、提取时间、提取次数,并以此优化工艺进行试验,所得总多酚提取率为17.11%。[结论]采用热浸提工艺提取石榴皮总多酚并用响应面分析法优化试验,在保证快速、高效的同时也具有节能、不破坏有效成分的优势,以纯水作为浸提溶剂更是避免了溶剂残毒问题,极大地提高了其安全性,因此也拓宽了极具生物活性的石榴皮多酚在生产生活领域中的应用。 [Objective] To study the optimal conditions for extraction of total polyphenols from pomegranate peel and determine its content.[Method]With pomegranate peel as raw material,pure water as solvent,total polyphenols were extracted from pomegranate peel.The variation law of yield of total polyphenols with temperature,time,solid-liquid ratio and extraction times was explored by single factor test,the extraction process was optimized and perfected by response surface analysis,and content of total polyphenols in pomegranate peel was determined by Folin-Ciocalteu colorimetric method.[Result]The optimal conditions were as following:temperature 45 ℃,time 30 min,solid-liquid ratio1∶20(g∶mL),extraction times 3.The order of influence degree from high to low was temperature,solid-liquid ratio,time,extraction times.Under the above conditions,the yield of total polyphenols could up to 17.11%.[Conclusion] Using heat extraction and response surface method is not only fast,efficient but also energy-saving and undamaged.With pure water as extraction solvent can avoid the problem of residual hazard,greatly improve its security,and thus widen the highly bioactive pomegranate peel polyphenols applied in the field of production and living situation.
