摘要
[目的]本研究对森林草莓中组蛋白去乙酰化酶基因(HDAC)进行鉴定和分析,并研究其在生长发育以及逆境胁迫响应过程中的表达变化,为深入研究森林草莓组蛋白去乙酰化调控机制奠定基础。[方法]运用生物信息学方法,对森林草莓中HDAC基因进行鉴定,并进行蛋白结构域、染色体定位、基因复制类型和其他进化分析。利用转录组数据分析森林草莓中HDAC基因在不同组织和发育阶段中的表达变化。运用实时荧光定量PCR对森林草莓中HDAC基因在低温、高温胁迫下的表达变化进行分析。[结果]森林草莓中包含12个HDAC基因,其中RPD3/HDA1家族9个,SIR2家族2个,HD2家族1个;RPD3/HDA1家族基因可以分为5类,SIR2家族中基因分为2类。森林草莓中RPD3/HDA1和HD2家族HDAC基因主要通过离散型复制模式进行扩张,SIR2家族在双子叶进化过程中没有发生明显扩张。各个HDAC基因具有发育阶段或组织特异性表达。部分HDAC基因在森林草莓热胁迫处理中有不同程度的响应,对冷胁迫的反应相对较小。[结论]森林草莓HDAC基因具有植物的典型特性,并可能参与了森林草莓的生长发育和对热胁迫的响应。
[Objectives] This research aims to identify the histone deacetylase (HDAC)genes in Fragaria vesca,and explore their characteristics.We also analyzed the expression patterns of HDAC genes during development and in response to cold and heat stresses in Fragaria vesca.Our results will shed light on further studies on the regulatory mechanisms of histone acetylation in strawberry.[Methods] We used bioinformatic approaches to identify and characterize the HDAC genes in Fragaria vesca.Phylogenetic analysis,domain constructs,chromosomal locations and gene duplication events were analyzed in Fragaria vesca and other horticulture species as well.Transcriptome data were used to analyze the expression patterns of HDAC genes in different tissue of Fragaria vesca,qRT-PCR was used to explore the response of HDAC genes towards cold and heat stresses.[Results] Twelve HDAC genes were identified in Fragaria vesca,including 9 in RPD3/HDA1 family,2 in SIR2 family and 1 in HD2 family.Gene duplication analysis showed that the expansion of RPD3/HDA1 and HD2 families were mainly caused by dispersed duplication events,and that no obvious expansions occurred in SIR2 family in the dicotyledons investigated.Transcriptome data indicated that HDAC genes had tissue-and developmental-specific expressions.In addition,qPCR data showed that histone deacetylase genes also had different expression patterns in response to heat stress,while response to cold stress was relatively smaller.[Conclusions] FvHDAC has plant-specific characteristics and may be involved in development and stress responses in Fragaria vesca.
