注射促肾上腺皮质激素对母猪断奶后新生黄体的发育及类固醇合成的影响

摘要 [目的]本文旨在确定应激对母猪排卵后黄体组织的正常发育及其功能的影响。[方法]采用注射促肾上腺皮质激素(ACTH)的方式模拟应激状态,利用RT-q PCR和免疫组织化学法研究新生黄体的类固醇合成能力和组织发育状况。[结果]ACTH组母猪血浆中皮质醇含量明显高于对照组母猪,新生黄体内胆固醇侧链裂解酶P450(P450scc)和3β-羟基类固醇脱氢酶(3β-HSD)的表达量比对照组母猪明显下调;ACTH组母猪基质金属蛋白(MMP)和血管内皮生长因子(VEGFa)的表达量明显低于对照组母猪;组织学检测结果也显示ACTH组母猪的黄体组织填充率低于对照组母猪,而且黄体内血管密度相对较低。[结论]注射ACTH模拟应激,可诱导受试母猪血浆高皮质醇含量,而且能够显著降低新生黄体内主要孕酮合成酶的表达和黄体组织内部的血管新生。提示,应激能够影响母猪新生黄体的正常发育和功能。 [Objectives] The purpose of the present study was to evaluate the impact of development and steroidogenesis of newly formed corpus luteal(CL) by stress in sows.[Methods] After injection of adrenocorticotropic hormone(ACTH) for simulating stress physiologic response,the tissue developmental status and the ability of steroidogenesis of the newly formed CL were examined by the RT-qPCR and immunohistochemistric methods.[Results] The results showed that plasma cortisol level in the ACTH treated sows was elevated significantly compared with the control.The expressions of P450 cholesterol side chain cleavage (P450scc) and 3β-hydroxysteroid dehydrogenase (3β-HSD)were down-regulated in the new CL of ACTH treated sows.The expressions of MMPs and VEGFa in the CL decreased compared to those of control sows.The ovary sections revealed a lower filling rate of luteal cells,and the capillary density in the CL was reduction in the ACTH treated sows.[Conclusions] The results suggested that the expression of main progesterone synthases and the angiogenesis of the newly formed CL in weaned sows could be impaired by higher plasma cortisol level because of ACTH administration.Stress can impact the normal development and function of the newly formed CL in sows.
