摘要
目的:探讨糖皮质激素治疗对重症肌无力患儿辅助性T1细胞(helper T cell 1,Th1)与调节性T细胞(regulatory cells,Tregs)转录及相关因子表达的影响。方法:160例重症肌无力患儿,分为观察组与对照组两组,同时取60名健康儿童作为健康组。观察组进行糖皮质激素治疗,对照组不进行糖皮质激素治疗,测定三组患儿的叉头转录因子(forkhead transcription factor p3,FOXP3)、T-bet(T-box expressed in T cells)因子的信使核糖核酸(messenger ribonucleic acid,mRNA)含量、转化生长因子-β1(transforming growth factor-β1,TGF-β1)、白细胞介素-6(interleukin-6,IL-6)、γ-干扰素(Interferon-γ,IFN-γ)的浓度并进行对比。结果:观察组患儿的T-bet mRNA、FOXP3 mRNA表达水平均高于对照组与健康组患儿(P<0.05);对照组患儿的血清TGF-β1、IL-6和IFN-γ的浓度高于观察组与健康组,且TGF-β1浓度差距最为明显(P<0.05);观察组患儿的T-bet和FOXP3 mRNA的表达比值高于对照组与健康组(P<0.05)。结论:糖皮质激素治疗能够降低T-bet mRNA与FOXP3 mRNA的表达水平,提升患儿免疫功能。
Objective: To investigate the effect of glucocorticoid therapy on the expression of Thl/Treg cells transcription factors and related cytokines in children with myasthenia gravis. Methods: 160 children with myasthenia gravis were divided into the observation group and the control group, at the same time, 60 healthy children were selected as the healthy group. The observation group was treated with glucocorticoid, while the control group was not treated with glucocorticoid. The concentration of forkhead transcription factor p3 ( FOXP3 ) mRNA, T - box expressed in T ceils ( T - bet ) mRNA, transforming growth factor - β1 ( TGF - β1 ), interleukin - 6 ( IL - 6 ) and Interferon - 3' ( IFN - 3' ) in the children of the three groups was measured and compared. Results : The expression of T - bet mRNA and FOXP3 mRNA in the observation group was higher than that in the control group and the healthy group( P 〈0.05). The concentration ofTGF - β1 ,IL -6 and IFN - γin the control group was higher than that in the observation group and the healthy group, and the concentration of TGF - 131 was the most obvious( P 〈 0.05 ). Conclusion: Glucocor- ticoid therapy can reduce the expression level of T - bet mRNA and FOXP3 mRNA and improve the immune function in children.
