摘要
目的:通过检测basigin在雌性去卵巢小鼠脑皮质及海马组织中的表达变化,探讨体内雌激素水平与脑组织basigin表达的关系。方法:将28只小鼠随机分为手术组和假手术组,每组14只。手术组小鼠经背侧行卵巢切除术;假手术组不摘除卵巢。2个月后采用免疫组织化学法(immunohistochemistry,IHC)和western blot技术检测小鼠脑部皮质和海马basigin的表达变化。结果:IHC结果显示,相对于假手术组,手术组小鼠脑皮质和海马的CA1、CA3区basigin表达水平降低(P<0.05);Western blot结果表明,手术组小鼠脑皮质和海马basigin表达水平低于假手术组(P<0.05)。结论:脑皮质及海马basigin的低表达与体内雌激素水平下降有关。
Objective:To investigate the relationship between the level of estrogen in the body and the expression of basigin in brain tissue by detecting the expres- sion changes of basigin in oophorectomy mice cortex and hippocampus. Methods: The 28 mice were randomly divided into operated and sham - operated groups for each group was contained 14 mice. The operated group mice were removed ovaries via dorsal oophorectomy; sham -operated group mice were not removed ovaries. Immunohistochemistry (IHC) and western blot were applied to observe the expression of basigin in cortex and hippecampus after two months. Results: Compared with sham -operated group, the IHC data showed the expression levels of basigin were declined both in cortex and the CA1, CA3 areas in hippocampus ( P 〈 0.05 ) ; the western blot data indicated that the protein levels were reduced in operated mice cortex and hippocampus, respectively ( P 〈 0.05 ). Condusion: Lower expression of basiginin cortex and hippocampus was related to the decline of estrogen levelin body.
