摘要
目的:观察精氨酸酶抑制剂(N-hydroxy-nor-L-a rginine,nor-NOHA)对大鼠心肌缺血再灌注损伤(myocardial ischemia reperfusion injury,MIRI)的保护作用。方法:将SD大鼠随机分成模型组和治疗组,采用结扎左冠状动脉前降支法建立心肌缺血再灌注模型。治疗组大鼠在造模之前给予nor-NOHA腹腔注射(20 mg/kg,1次/d),连续注射2周。比较两组大鼠心肌梗死面积占左室面积比以及心肌精氨酸酶表达水平。结果:MIRI后2 h,治疗组大鼠心肌梗死面积占左室面积比小于模型组(t=2.133,P=0.044),MIRI后24 h,治疗组大鼠心肌梗死面积占左室面积比小于模型组(t=3.171,P=0.006);MIRI后2 h,治疗组大鼠心肌精氨酸酶水平低于模型组(t=6.117,P=0.002)。结论:norNOHA对MIRI大鼠心肌精氨酸酶的表达有抑制作用,nor-NOHA能在一定程度上缩小MIRI心肌坏死的面积。
Objective: To observe the theprotective effect ofN - hydroxy - nor - L - a rginine ( nor - NOHA ) on myocardial isehemiareperfusion injury (MIRI) in rats. Methods:The SD rats were randomly divided into the model group and the treatment group. Myocardial ischemia reperfusion model were established by ligation of left anterior descending coronary artery. The rats in the treatment group were given intraperitoneal injection of nor - NOHA ( 20 mg/kg, twice / day) before modeling, and were injected for 2 weeks. The ratios of myocardial infarction areas to the left ventric- ular areasand the expression levels of the cardiac muselewere compared in the two groups. Results: 2 h after the treatment of myocardial infarction area by MIRI, the ratio of myocardial infarction area to the left ventricnlar area in the treatment group was lower than that in the model group ( t = 2. 133, P = 0. 044 ) ; 24 h after the treatment of myocardial infarction areaby MIRI, the ratio of myocardial infarction area to the left ventricular area in the treatment group was lower than that in the model group ( t =3. 171, P =0.006) ; 2 h after the treatment of myocardial infarction area by MIRI, the arginase level of the treatment group was lower than that of the model group( t = 6.117, P = 0. 002 ). Conclusion : Nor - NOHA has an inhibiting effect on the expression levels of the cardiac muscleand can reduce infarct size caused by MIRI.
