摘要
目的:对益气化瘀清热方进行拆方,通过观察各类含药血清对脂多糖(lipopolysaccharide,LPS)诱导下体外培养的大鼠肾小球系膜细胞(primary mesangial cells,Ms C)表达TGF-β1及Smad3蛋白的影响,探讨不同类中药的作用强度以及益气化瘀清热方治疗肾小球肾炎的作用机制。方法:采用Wersten Blot法检测各组含药血清对大鼠Ms C表达TGF-β1及Smad3蛋白的影响。结果:Wersten印迹结果显示:各组含药血清均能抑制体外培养的Ms C表达TGF-β1及Smad3蛋白,与对照组比较,差异有统计学意义(P<0.05);其中复方组作用最强,与其他含药血清组比较差异有统计学意义(P<0.05),清热组与化瘀组比较无明显差异(P>0.05),但优于益气组,差异有统计学意义(P<0.05)。结论:益气化瘀清热方及其拆方、雷公藤多苷均能抑制体外培养的大鼠Ms C表达TGF-β1及Smad3蛋白的表达。复方组作用明显优于其他组,清热组及化瘀组次之,益气组最弱。
Objective: Herbs in Yiqi Huayu Qingre Decoction were divided into different groups according to their unique efficacy and then the influence of medicated serum to the expression proteins of TGF-β1 and Smad3 in rat mesangial cells which were induced by lipopolysaccharide and cultured in vitro were observed,through which the intensity of the action of these herbs,along with the mechanism of the entire decoction treating glomerulonephritis were explored.Methods: The effect of medicated serum to the Ms C expressing proteins of TGF-β1 and Smad3 was observed by Wersten Blot. Results: The results showed all kinds of medicated serum can restrain the expressions of TGF-β1 and protein Smad3 in rat mesangial cells and there was a statistic significance compared with control group( P〈0. 05),among which,compound group had the strongest effect. In addition,there was no obvious difference in clearing heat drugs group and resolving stasis drugs group( P〉0. 05). The difference was much evident when compared to benefiting Qi group. Conclusion: Yiqi Huayu Qingre Decoction and its disassembled prescriptions as well as TW all can restrain the expressions of TGF-β1 and protein Smad3 in rat mesangial cells cultured in vitro. However,the compound group was better than the others while the effects of clearing heat group and resolving stasis group comes second and benefiting Qi group has the weakest effect.
