大黄对便秘大鼠肠动力及结肠水通道蛋白3表达的调节作用 被引量:22

摘要 目的:采用洛哌丁胺灌胃建立便秘大鼠模型,观察大黄对便秘模型大鼠AQP3表达的影响。方法:健康Wistar大鼠18只,随机分为3组,空白组、模型组、大黄组。模型组及大黄组采用配制好的含洛哌丁胺(1.5 mg·kg^(-1)·d^(-1))混悬液2 m L灌胃7 d。模型建立后,大黄组以大黄(5 mg·kg^(-1))混悬液2 m L灌胃。检测结肠传输,采用RT-PCR、免疫组织化学法检测AQP3表达情况。结果:(1)造模后大鼠的体重增加、结肠存留粪便增多、粪便含水量降低、肠道传输时间延长(P<0.05)。(2)大黄干预组大鼠大便量、粪便含水量增加(P<0.05)。(3)大黄干预便秘大鼠,其结肠传输时间缩短,AQP3 mRNA及蛋白表达水平低于模型组(P<0.05)。结论:大黄能抑制AQP3在大鼠结肠黏膜层的表达,其表达降低可能与下调结肠上皮细胞质膜上功能性AQP3数量相关,从而增加粪便含水量。 Objective: To observe the effect of Rhubarb on expression of Aquaporin-3 mRNA in colon tissues in the Lopersmide-induced constipated rats. Methods: Eighteen rats with constipation by irrigating the rats' stomach with Lopersmide were set up and divided into 3 groups: control group,model group and Rhubarb group at random. Model group and Rhubarb group were used to choose the fittest dosage of Lopersmide( 1. 5 mg· kg^-1·d^-1) by irrigating the rats' stomach to establish the constipation model during seven days. After the establishment of the model,Rhubarb( 5 mg·kg^-1) was given by intragastric administration in Rhubarb group. Intestinal transmission function was determined. RT-PCR and immunohistochemistry were used to examine the expression of AQP3. Result:(1)There was significant increase in the aspect of body weight,feces in the colon and intestinal transit time whereas fecal water content was remarkable reduced in model rats( P〈0. 05).(2)The fecal water content was increased obviously in Rhubarb group( P〈0. 05).(3)Rhubarb could obviously accelerate intestinal transit and depress the expression of AQP3 in colon( P〈0. 05). Conclusions: Rhubarb can depress the expression of AQP3 in rats with constipation of colon tissues. It may be correlated to decreasing the functional AQP3 quantity and increased the fecal water content.
