摘要
目的:探讨莪术油对阿霉素诱导耐药的人甲状腺未分化癌细胞株HTh74Rdox增殖和耐药性的影响及机制。方法:用不同浓度莪术油处理HTh74Rdox后观察细胞形态;MTT检测莪术油对HTh74Rdox细胞的生长抑制作用;流式细胞术检测HTh74Rdox细胞凋亡;Western Blot检测Bcl-2和Bax蛋白的表达;q PCR检测MDR1和ABCG2 mRNA的变化。结果:莪术油对HTh74Rdox细胞的增殖有明显抑制作用,联合阿霉素后可减少阿霉素剂量;莪术油明显诱导细胞凋亡(P<0.01);且能够提高Bax/Bcl-2比值(P<0.05),降低MDR1、ABCG2 mRNA表达。结论:莪术油可以抑制HTh74Rdox细胞的生长并改善其耐药作用,其机制可能分别与提高Bax/Bcl-2比值和降低MDR1、ABCG2 mRNA表达有关。
Objectives: To explore the influence of zedoary turmeric oil in proliferation and drug resistance of HTh74 Rdox cells and producation. Methods: HTh74 Rdox cells were to be treated by different concentrations of Zedoary turmeric oil to observe the morphological change and apoptosis situation of HTh74 Rdox cells. MTT was used to test inhibiting effect of Zedoary turmeric oil on the proliferation of HTh74 Rdox cells. The cell cycle change and cell apoptosis rate were further detected by flow cytometry. Western Blot was used to measure the expressions of Bcl-2 and Bax. And finally,with the help of q PCR,the mRNA expressions of MDR1 and ABCG2 were detected. Results: Zedoary oil played an significant role in inhibiting the growth of HTh74 Rdox cells. Combination of zedoary turmeric oil and adriamycin could reduce the average dose for Adriamycin. Zedoary turmeric oil significantly increased the percentage of Bax/Bcl-2 and reduced the mRNA expressions of MDR1 and ABCG2. Conclusions: Zedoary turmeric oil can inhibit the growth of HTh74 Rdox cells and improve its drug resistance significantly. And the possible mechanisms for zedoary turmeric oil are related to the increase of Bax/Bcl-2 ratio and the reduction of MDR1 and ABCG2 mRNA expressions respectively.
