脾肾气虚证2型糖尿病大鼠模型的复制 被引量:5

摘要 目的:观察通过饮食劳倦所伤复合腹腔注射丁胺卡那霉素及链脲佐菌素作为造模要素能否成功复制出脾肾气虚证2型糖尿病病证结合动物模型。方法:健康雄性SD大鼠60只,体质量(180±20)g,按体质量随机分成二组:空白对照组(n=20)和模型组(n=40)。模型组大鼠单日用200%的大黄浓煎液灌胃,4 m L/只,游泳至耐力极限;双日高糖高脂膳食持续喂养,共计12周,第4周后给予丁胺卡那霉素及链脲佐菌素腹腔注射。结果:模型组大鼠造模4周后出现游泳耐力逐日下降,体毛光亮度减退,倦怠,懒动,喜聚堆、弓背蜷缩,粪便时软、时塘,血糖增高;第6周后体质量开始显著增加;甘油三酯和高密度脂蛋白分别从第2周后表现出异常;实验结束后模型组出现高胰岛素血症、胰岛素抵抗,符合本研究拟建立的脾肾气虚证、2型糖尿病的评估标准。结论:通过饮食劳倦所伤复合腹腔注射丁胺卡那霉素及链脲佐菌素造模方法可成功复制出脾肾气虚证2型糖尿病病证结合动物模型。 Objective: We observed that whether the modeling approach by fatigue plus improper diet and intraperitoneal injection of amikacin and streptozotocincan can copy deficiency syndrome T2 DM rat model. Methods: Sixty healthy male Sprague Dawley rats,weighing( 180 ± 20) g,were randomly divided into two groups: blank control group( n = 20)and model group( n = 40). The model group were gavaged with 200% rhubarb concentrated decoction,4 m L/rat and swimmed to the endurance limit in a single day. The model group were fed high sugar,high fat diet feeding in a double day,a total of 12 weeks. Use intraperitoneal injection of amikacin and streptozotocincan after 4th week. Results: Rats model showed swimming endurance was decreased,the brightness of animal hair loss,fatigue,lazy,like the heap,arched back and curled up,abnormal stool,increased blood sugar. The weight began to increase significantly after 6 weeks. Triglycerides and high density lipoprotein were abnormal after 2 weeks. Rats showed high insulin resistance and insulin resistance with spleen and kidney deficiency syndrome. Conclusion: The modeling approach by fatigue plus improper diet and intraperitoneal injection of streptozotocincan can copy spleen and kidney deficiency syndrome of T2 DM rat model.
