Expression and Clinical Significance of Stromal Cell Derived Factor-1 and Nerve Growth Factor in Non-small Cell Lung Cancer

摘要 目的:探讨间质细胞衍生因子-1(SDF-1)与神经生长因子(NGF)在非小细胞肺癌(NSCLC)中的表达及临床意义。方法:收集2014年2月-2016年5月本院胸外科手术切除的人NSCLC病理标本65例作为观察组,其中观察组按不同病理级别分为了Ⅰ级15例为Ⅰ组,Ⅱ级24例为Ⅱ组,Ⅲ级26例为Ⅲ组。同时随机选择健康者正常肺组织标本64例为对照组,各组组织均使用实时荧光定量聚合酶链式反应(RT-PCR)技术检测SDF-1与NGF mRNA,同时采用免疫组化法检测各组组织中SDF-1与NGF蛋白的表达情况。并分析比较各组组织中SDF-1与NGF mRNA及蛋白表达情况。结果:观察组SDF-1与NGF mRNA基因表达量及蛋白阳性率均高于对照组(P<0.05);Ⅲ组SDF-1与NGF mRNA基因表达量及蛋白阳性率均高于Ⅱ组(P<0.05);Ⅱ组SDF-1与NGF mRNA基因表达量及蛋白阳性率均高于Ⅰ组(P<0.05);NSCLC组织中SDF-1与NGF mRNA基因表达量及蛋白阳性率随病理级别的增高而增高(P<0.05)。SDF-1和NGF高表达均是预测NSCLC预后不良的独立因素(P<0.01),SDF-1和NGF的高表达与NSCLC患者整体不良预后显著相关(P<0.01)。结论:NSCLC中SDF-1与NGF高表达对病变进展有一定促进作用,两者与肿瘤的病理级别密切相关,SDF-1与NGF在NSCLC的发生发展中扮演重要的角色,有望为NSCLC的临床检测及治疗提供新思路。 Objective: To investigate the expression and clinical significance of stromal cell derived factor-1 ( SDF-1 ) and nerve growth factor ( NGF ) in non-small cell lung cancer ( NSCLC ) .Method: 65 NSCLC patients who underwent surgical resection in our hospital from February 2014 to May 2016 were selected as observation group, the observation group was divided into different groups according to different pathological grades, 15 cases in grade Ⅰ as the group Ⅰ, 24 cases in grade Ⅱ as the group Ⅱ, 26 cases in grade m as the group Ⅲ.At the same time, randomly selected healthy subjects of normal lung tissue specimens of 64 cases for the control group.All groups were used real-time fluorescence quantitative polymerase chain reaction ( RT- PCR ) technique to detect SDF-1 and NGF expression of mRNA, and immunohistochemical detected SDF- 1 and NGF protein in tissue, and tissues were analyzed and compared with NGF SDF-1 mRNA and protein expression.Result: The SDF-1 and NGF mRNA gene expression and protein positive rates in observation group were significantly higher than those in control group ( P〈0.05 ) ; the SDF-1 and NGF mRNA gene expression and protein positive rates in group Ⅲ were higher than those of group Ⅱ ( P〈0.05 ) ; the SDF-1 and NGF mRNA gene expression and protein positive rates in group Ⅱ were higher than those of group Ⅰ( P〈0.05 ) ; the expression of SDF-1 and NGF in NSCLC tissues and the positive rate of mRNA increased with the increase of pathological grade ( P〈0.05 ) .The high expression of SDF-1 and NGF were independent predictors of poor prognosis of NSCLC ( P〈0.01 ), and the high expression of SDF-1 and NGF were significantly correlated with the poor prognosis of patients with NSCLC (P〈0.01) .Conclusion: The high expression of SDF-1 and NGF in NSCLC may promote the progression of the disease, both are closely related to the pathological grade of the tumor, SDF-1 and NGF play an important role in the occurrence and development of NSCLC, which is expected to provide new ideas for clinical detection and treatment of NSCLC.
