美洲大蠊提取物对大鼠血液和卵巢抗氧化指标的影响 被引量:5

摘要 为了研究美洲大蠊提取物是否对大鼠血液和卵巢抗氧化指标产生影响,试验使用成年雌性SD大鼠17只,适应饲养1周后,随机分为3组,对照组(9 m L/kg,生理盐水)5只、低剂量组(9 m L/kg,0.03 g/m L)6只、高剂量组(9 m L/kg,0.09 g/m L)6只,隔天灌胃1次,共饲养30 d。试验结束后采集大鼠血液和卵巢检测血清丙二醛(MDA)、一氧化氮(NO)含量和超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px)活性以及卵巢SOD、GSH-Px、CAT基因表达的变化。结果表明:与对照组相比,高剂量组(P=0.085)、低剂量组(P=0.091)均有显著降低血清中NO含量的趋势;高剂量和低剂量的美洲大蠊提取物均显著升高了大鼠血清中SOD的活性(P<0.05),同时高剂量组还显著降低了血清中MDA的含量(P<0.05);此外,高剂量和低剂量组还显著升高了卵巢SOD基因的表达(P<0.05)。美洲大蠊提取物能升高大鼠血清抗氧化指标SOD的活性,降低了氧化应激指标NO和MDA的含量,同时还升高了卵巢SOD基因的表达量,说明其对大鼠卵巢具有一定的抗氧化应激功能。 To study whether Periplaneta americana extract had.an effect on the antioxidant indexes of blood and ovary of rats, 17 adult female SD rats were used in the test. The rats were randomly divided into three groups after a week of feeding adaption, inkling a control group of 5 rats administrated with physiological saline of 9 mL/kg, a low - dose group of 6 rats administrated with physiological saline of 9 mlJkg and Periplaneta americana extract of 0.03g/mL, and a high - dose group of 6 rats administrated with physiological saline of 9 mL/kg and Periplaneta americana extract of 0.09 g/mL. All rats were administrated with intragastric gavage once every other day, and reared for 30 d. The blood and ovaries of rats were collected after the test, and the content of malondialdehyde ( MDA), nitric oxi'de(NO) level, the activities of superoxide dis- mutase (SOD), catalase (CAT), glutathione perexidase (GSH -Px) in the serum, the changes in the gene expressions of SOD, GSH -Px and CAT in the ovary were detected, respectively. The results showed that the high dose group ( P = 0. 085 ) and low dose group ( P = 0. 091 ) had a significant trend to reduce NO level in serum of rats compared with control group. High dose and low dose of Periplaneta americana extracts significantly increased (P 〈 0.05 ) the SOD activity in serum of rats. However, the high dose group significantly decreased (P 〈 0.05 ) the content of MDA in serum of rats. In addition, the high dose and low dose groups significantly increased ( P 〈 0.05 ) the SOD gene expression in the ovary. Periplaneta americana extract increased the SOD activity as the antioxidant index in the serum of rats and reduced the oxidative stress indicators of NO and MDA contents. It also increased the expression level of SOD gene in the ovary of rats. The results indicate that Periplaneta americana extract has a certain antioxidant stress function to the ovary of rats.
