典型中性蛋白酶制剂的活性酶蛋白组分的分离及其对胶原蛋白的作用性能 被引量:3

摘要 用Superdex 75 prep grade凝胶过滤色谱对三种典型中性蛋白酶制剂进行了分离纯化,得到了具有不同相对分子质量的蛋白组分;用SDS-PAGE电泳测定了活性酶蛋白的相对分子质量,并对其酪蛋白、胶原蛋白和弹性蛋白的作用性能进行了评价;考察了三种酶活力组分的脱毛效果及其对皮粒面的作用程度。结果表明,三种中性蛋白酶制剂中的活性酶蛋白的相对分子质量不同,均无弹性蛋白水解活力,但对胶原蛋白的作用性能存在较大差异;酶的脱毛效果随其胶原活力的增加而加强,但皮革粒面的损伤程度随蛋白酶的胶原水解活力和相对分子质量的增加而加剧。 Three typical neutral protease preparations were purified through gel filtration chromatographywith the Superdex 75 prep grade column, and the proteins with different molecular weight were separated. The molecular weight values of the obtained active enzyme proteins were determined by SDS-PAGE eleetrophoresis, and their activities toward casein, collagen and elastin were also evaluated. Finally, the effects of the three enzyme activity components on hair-removal and grain-damage were investigated.The results show that the molecular weights of the three active enzyme proteins are different, and theyhave no elastinactivity,but exhibitdifferentproteolytieperformanee toward collagen;the protease with high collagen activity has better hair-removaleffeet, but the degree of grain-damage effect is aggravated with the increase of collagen activity and molecular weight of the active protein.
