白介素17A对口腔黏膜上皮细胞抗念珠菌黏附及β-防御素-2表达的影响 被引量:1

摘要 目的:探讨白介素17A(IL-17A)对白色念珠菌Candida albicans(C.a)感染的人口腔黏膜上皮细胞(OMECs)β-防御素-2(hBD-2)表达的影响。方法:将OMECs分为空白对照组、单独重组人IL-17A(rhIL-17A)处理组、单独C.a处理组(模型组)及C.a与rhIL-17A共同处理组(实验组)。按菌∶细胞=1∶10建立C.a感染的OMECs模型。分别将单独rhIL-17A处理组和实验组的OMECs按所加入的rhIL-17A浓度分为4个亚组:1μg/L组、10μg/L组、100μg/L组及1 000μg/L组,共培养48h。收集不同时间点(0h、12h、24h、48h)的细胞培养上清液,计算C.a黏附率;分别采用荧光定量PCR(FQ-PCR)和酶联免疫吸附试验(ELISA)方法检测hBD-2mRNA相对表达量及蛋白水平。结果:与模型组比较,实验组C.a黏附率有所降低,但差异无统计学意义(P>0.05)。不同浓度的rhIL-17A均能诱导OMECs表达hBD-2,且100μg/L组hBD-2mRNA相对表达量及蛋白水平升高最为显著(P<0.01)。采用100μg/L rhIL-17A刺激OMECs 12h、24h、48h后,hBD-2mRNA及蛋白水平均升高,且48h时hBD-2mRNA及蛋白水平显著高于0h和12h(P<0.05或P<0.01)。结论:IL-17A能诱导OMECs表达hBD-2,但对口腔黏膜上皮抗C.a黏附无明显作用。 Objective:To investigate the effect of interleukin 17A (IL-17A) on the expression of ^-defensin-2 (hBD^2) and the adherence of C. albicans to human oral mucosal epithelial cells (OMECs). Methods: OMECs was divided into 4 groups: a control group, an only recombinant human IL-17A (rhIL-17A) intervention group, a model group in which the ceils were infected with C. albicans (1 :10 ratio), and an experimental group in which the cells were infected with C. albicans and treated with different concentrations (1, 10, 100 and 1,000 μg/L) of rhIL-17A. The mRNA and protein levels of hBD-2 were detected by fluorescent quantitative PCR (FQ-PCR) and enzyme-linked immuno sorbent assay (ELISA), respectively. Results:C. albicans adherence rate in the experimental group was a little lower than that in the model group ( P 〉0.05). Different concentrations of rhIL-17A could up-regulate hBD-2 expression in OMECs, and 100 μg/L was the highest ( P 〈0. 01). Treated with 100 μg/L rhIL-17A for 12 h, 24 h and 48 h lead to markedly increased hBD-2 mRNA and protein levels in OMECs, but 48 h was the highest ( P 〈0.05 or P 〈0.01). Conclusion:IL-17A could induce the hBD-2 expression in OMECs, whereas had no effect on the adherence of C. albicans to OMECs.
