烟草烟雾暴露小鼠中CD4^+ Foxp3^+调节性T细胞亚型与Th1/Th17变化的实验研究 被引量:2

摘要 目的:探讨烟草烟雾暴露后小鼠CD4^+Foxp3^+调节性T细胞(Treg细胞)亚型及Th1、Th17细胞比例的变化及其意义。方法:将20只雄性Balb/c小鼠随机分为对照组和烟熏组,烟熏组小鼠烟草烟雾暴露24周建立肺气肿模型。采用苏木精—伊红(HE)染色法观察小鼠肺组织形态学改变。流式细胞术检测小鼠外周血、脾脏和肺组织中Treg细胞亚型及Th1、Th17的比例变化。结果:烟熏组肺泡平均内衬间隔(MLI)为(53.3±8.9)μm,明显高于对照组的(28.0±1.2)μm(P<0.01)。与对照组比较,烟熏组外周血、脾脏和肺组织中Th1、Th17细胞比例均升高(P<0.05),记忆型Treg细胞(CD4Foxp3^+CD44^+CD62^+)、效应型Treg细胞(CD4Foxp3^+CD44^+CD62-)比例升高,幼稚型Treg细胞(CD4Foxp3^+CD44^-CD62^+)比例下降,差异均有统计学意义(P<0.05)。结论:烟草烟雾暴露肺气肿小鼠体内Treg细胞亚型参与了气道炎症免疫调节,可能有助于减少Th1、Th17细胞的表达并减轻肺部炎症反应。 Objective: To explore the change of Treg cells subtype and Th1/Th17 cell proportion and the possible significance in a cigarette smoke-induced mice emphysema model. Methods: 20 male Balb/c mice were randomly divided into two groups: a control group and a smoke-exposure group, with 10 mice in each group. Alveolar airspace enlargement was observed by hematoxylin-eosin (HE) staining. Morphological change was evaluated by mean linear intercepts (MLI). The proportion of Thl/Th17 and Treg cells subtypes in peripheral blood, spleen and lungs of mice were determined by flowcytometry. Results: The MLI in smoke-exposure group was higher than that in the control group[(53.3±8.9) μm vs. (28.0±1.2) him, P 〈0.01)]. The ratio of Thl, Th17, memory Treg cells (CD4 Foxp3+ CD44+ CD62+) and effector Treg cells (CD4 Foxp3+ CD44+ CD62-) in peripheral blood, spleen and lungs of the smoke-exposure group were higher than those of the control group ( P 〈0.05). The naive Treg cells (CD4 Foxp3+ CD44 CD62+) in peripheral blood, spleen and lungs of the smoke-exposure group were slightly fewer than those in the control group ( P 〈0.05). Conclusion: Treg cells subtypes participated in airway inflammation and immune regulation of cigarette smoke-exposed mice. It may help reduce Thl and Th17 cells, and alleviate inflammatory response in the lungs.
