参芪扶正注射液对人前列腺癌PC-3细胞增殖的影响及机制研究

Antiproliferation Activity and Mechanism of Shenqi Fuzheng Injection(参芪扶正注射液) on Human Prostate PC-3 Cells

目的:研究参芪扶正注射液在体外对人前列腺癌PC-3细胞增殖的影响及其机制。方法:体外培养PC-3细胞,MTT和SRB法检测细胞增殖抑制率;PI染色法测PC-3细胞凋亡率;ELISA法测Caspase-3、Caspase-9、Fas含量;Bradford法测SOD、MDA含量。结果:参芪扶正注射液对PC-3细胞的体外增殖具有抑制作用,并具有浓度依赖性;参芪扶正注射液能诱导PC-3细胞凋亡,促进Caspase-3、Caspase-9、Fas蛋白表达,增加细胞内SOD的活性,降低MDA的含量。结论:参芪扶正注射液可抑制PC-3细胞的增殖,其作用机制与启动线粒体通路、死亡受体通路和内质网通路及增加机体的抗氧化功能有关。

Objective： To study the antiproliferation activity and mechanism of Shenqi Fuzheng injection on human prostate cancer PC-3 cells. Methods： PC-3 cells were cultured in vitro; MTT and SRB were performed to observe the proliferation. PI was utilized for measure the apoptosis. The expressions of Caspase-3, Caspase-9 and Fas were determined by ELISA. The activity of SOD and content of MDA were assayed with corresponding kits respectively. Results： Shenqi Fuzheng injection could inhibit the proliferation of PC-3 cells in vitro, which was concentration-dependent, PC-3 cells were induced apoptosis, the expressions of Caspase-3, Caspase-9 and Fas were promoted, the activity of SOD increased, MDA decreased significantly. Conclusion： Shenqi Fuzheng injection inhibits the proliferation of PC-3 cells, which may related with start the mitochondrial pathway, death receptor pathway, endoplasmic reticulum road, and increase the antioxidant function.
