p38MAPK信号通路调控IL-6、HIF-1α及VEGF对慢性鼻-鼻窦炎发病机制的影响 被引量:8

摘要 目的观察p38丝裂原活化蛋白激酶(p38MAPK)在慢性鼻-鼻窦炎(CRS)不伴鼻息肉(CRSsNP)、CRS伴鼻息肉(CRSwNP)以及正常钩突黏膜组织中表达程度的差异以及不同类型的CRS中细胞因子白介素6(IL-6)、缺氧诱导因子-1α(HIF-1α)和血管内皮生长因子(VEGF)的表达,探讨并分析p38MAPK信号通路在CRS中可能的作用机制。方法选取2015年7月~2016年7月青岛市市立医院耳鼻喉科行鼻内镜手术患者,32例CRSsNP患者窦口组织、30例CRSwNP患者鼻息肉组织和24例正常钩突黏膜组织,分别采用蛋白质印迹法(Western Blot)检测p38MAPK在三组中的蛋白表达量、用逆转录聚合酶链式反应法(RT-PCR)检测IL-6、HIF-1α和VEGF在三组中的信使核糖核酸(mRNA)的表达水平。结果 p38MAPK在CRSsNP、CRSwNP中的表达均高于正常黏膜组织,而两组实验组间的表达差异无统计学意义(P=0.143);IL-6在CRSsNP、CRSwNP中的表达均高于正常黏膜组织,而两组实验组间的表达差异无统计学意义(P=0.082);HIF-1α和VEGF在CRSwNP中表达均明显高于正常组织(P<0.05),而VEGF在CRSsNP组织和正常组织中的表达差异无统计学意义(P=0.075)。结论 p38MAPK在CRSsNP和CRSwNP中呈现出与炎性因子IL-6不同的相关性;IL-6在CRSsNP和CRSwNP中表达优势不同,在CRSsNP组织中表达量更显著,与其关系更密切;HIF-1a及VEGF在CRSwNP组织中同时呈高表达,可能与息肉的产生相关。 Objective To observe the difference of the degree of expression of p38 mitogen-activated protein kinase (p38 MAPK) in chronic rhinosinusitis (CRS) without nasal polyps (CRSsNP),CRS with nasal polyps (CRSwNP) and normal uncinate process,and the expression of interleukin-6 (IL-6),hypoxia-inducible factor 1 alpha (HIF-1α) and vascular endothelial growth factors (VEGF) in different types of CRS.And explore the possible mechanism of p38MAPK signaling pathway in CRS.nethods Patients with nasal endoscopic surgery in department of otolaryngology of Qingdao Municipal Hospital were selected from July 2015 to July 2016 in Qingdao municipal hospital otolaryngology.32 cases of patients with CRSsNP sinus tissue,30 cases of CRSwNP patients with nasal polyps and 24 cases of with normal uncinate mucosa were selected for the study.The protein expression of p38 MAPK in three groups was detected by Western blotting.And the mRNA expression levels of IL-6, HIF-1α and VEGF in three groups were detected by reverse transcriptase-polymerase chain reaction (RT-PCR). Results The expression of p38MAPK in CRSsNP and CRSwNP was higher than that in normal mucosa, but there was no significant difference between the two groups (P=0.143).The expression of IL-6 in CRSsNP and CRSwNP was higher than that in normal mucosa,but there was no significant difference between two groups (P=0.082).The expression of HIF-1α and VEGF in CRSwNP was significantly higher than that in normal tissues (P〈0.05).While there was no significant difference in the expression of VEGF in CRSsNP and normal tissues (P=0.075).Conclusion p38 MAPK in CRSsNP and CRSwNP shows a different correlation with inflammatory factor IL-6,and the advantage of the expression of IL-6 in CRSsNP and CRSwNP is different,and the expression of IL-6 in CRSsNP is more significant and more closely related.HIF-la and VEGF are also highly expressed in CRSwNP tissues,which may be related to the production of polyps.
