葛根异黄酮降血糖活性及作用机制的研究 被引量:36

摘要 目的:探讨葛根异黄酮降血糖活性及作用机制。方法:一次性注射150 mg/kg链脲佐菌素(STZ),建立糖尿病小鼠模型,动物分为空白组、损伤组、阳性对照组(消渴丸)、葛根异黄酮保护组,采用连续30 d灌胃给药法,通过测定血糖(BG)、体重、脏器指数、血清中总胆固醇(TC)、甘油三酯(TG)含量、肝脏中丙二醛(MDA)、蛋白羰基化(PCO)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)含量,研究STZ对小鼠模型的损伤程度及葛根异黄酮的保护作用。结果:葛根异黄酮能够显著降低模型血糖,改善小鼠体重下降的趋势,降低血清中TC、TG的含量(P<0.01,P<0.05),增强肝脏中SOD、CAT活性(P<0.05),降低肝脏中MDA、PCO含量(P<0.05)。结论:葛根异黄酮能够增强糖尿病小鼠的抗氧化能力,改善糖尿病小鼠的糖脂代谢,对糖尿病小鼠有较好的保护作用。 Objective: This paper is to investigate the protective effect of Radix Puerariae isoflavone on diabetic mice. Methods: We established type Ⅱ diabetes mouse model of by injecting streptozotocin(STZ) one-time. The mice were divided into blank control group, injury group, positive control group(xiaokewan) and Radix Puerariae isoflavone group, all of which used continuous 30 d gavage method. In order to determine the damage of STZ and the protective effect of Radix Puerariae isoflavone on type Ⅱ diabetes mice, we detected the content of glucose, body weight, organ index, serum biochemical indexes including total cholesterol(TC) and triglyceride(TG), antioxidant enzymes superoxide dismutase(SOD), catalase(CAT) and oxidative damage enzymes malondialdehyde(MDA), protein carbonyl(PCO). Results: The extraction of Radix Puerariae isoflavone significantly reduced the model of glucose, improved the decreasing body weight, and reduced the content of TC and TG in serum of mice(P〈0.01, P〈0.05),meanwhile, it enhanced SOD activities and reduced the MDA content in liver(p〈0.05). Conclusion: Therefore, Radix Puerariae isoflavone enhanced the antioxidant capacity, and improved glucose and lipid metabolism on diabetic mice, which showed a better protective effect on diabetic mice.
