摘要
以八月中旬在五峰采集的白芨未成熟种子为外植体,通过植物组织培养技术,对白芨进行无菌播种,后通过丛生芽诱导及生根培养,以期得到的白芨组培苗,并对其进行炼苗。实验研究了植物生长调节剂对白芨无菌萌发率及其成苗的影响,总结了白芨各个生长阶段最佳的培养基组分。实验表明:白芨种子萌发最适培养基为:1/2 MS+1 mg/L NAA+2 mg/L 6-BA+3%蔗糖+0.7%琼脂,接种20 d后的种子萌发率为98%,萌发时间快而集中,出芽整齐一致;白芨丛生芽诱导最适培养基为:1/2 MS+1 mg/L NAA+1 mg/L GA3+0.2%AC+3%蔗糖+0.7%琼脂,接种20 d后白芨丛生芽诱导率为75%,苗基部产生丛生芽量较多,生长较快;白芨生根最适培养基为:MS+0.2 mg/L NAA+3%蔗糖+0.8%琼脂,接种20 d后白芨生根率为0.95%,苗根粗壮,长势好,且出苗整齐。该实验结果为白芨规模化生产提供了一定依据,在白芨资源利用和异地保护上具有重要意义。
This study used the immature seeds ofBletilla striata that were collected from Wufeng in the middle of August as explants, and seeded them in asepic environment by plant tissue culture techniques. Then we used multiple shoot induction and rooting culture to get the tissue culture seedlings ofBletilla striata and let them adapt to the nature environment. The studies analyzed the influence of plant growth regulator on Bletilla striata aseptic germination rate and seedling, and summarized the best culture medium composition at each growth stage ofBletilla striata. The results indicated that the optimal medium for seed germination was 1/2 MS+1 mg/L NAA+2 mg/L 6-BA+3% sucrose+0.7% agar, and the germination rate was 98% after 20 d, which had fast and concentrated germination time, and uniform and consistent budding. The optimal medium for the induction of multiple shoot clumps was 1/2 MS+1 mg/L NAA+1 mg/L GA3 +0.2% AC+3% sucrose+0.7% agar, and the induction rate was 75% after 20 d. Moreover, multiple shoots had a large number and grew fast. The optimal medium medium for rooting ofBletilla striata was MS+0.2 mg/L NAA+3% sucrose+0.8% agar, and the rooting percentage is 0.95% after 20 d, with robust seedling roots, well growth and the orderly seedlings. The experimental results provided the basis for large-scale production ofBletillas striata, and had important significance in the utilization and protection of the resource of Bletilla s triata.
