肝癌相关抗原Cdc25C过表达树突状细胞疫苗的制备 被引量:1

摘要 目的制备肝癌相关抗原Cdc25C过表达树突状细胞疫苗,为研究Cdc25C在肝癌中的作用及机制奠定基础。方法构建Cdc25C过表达慢病毒载体,体外感染DC2.4细胞。同时以未感染的DC2.4细胞为空白对照,以感染慢病毒LV5空载体的DC2.4细胞为阴性对照。分别用倒置荧光显微镜观察各组DC2.4细胞绿色荧光蛋白的表达,用RT-PCR技术检测各组DC2.4细胞Cdc25C mRNA的表达,用Western blot技术检测各组DC2.4细胞Cdc25C蛋白的表达,用流式细胞术检测各组DC2.4细胞表面分子的表达。结果获得滴度为6×108TU/m L的Cdc25C慢病毒载体(LV5/Cdc25C),感染DC2.4细胞后,LV5/Cdc25C DC2.4组可见明显绿色荧光蛋白表达。RT-PCR和Western blot结果显示LV5/Cdc25C DC2.4组Cdc25C mRNA和蛋白的表达水平明显升高。经流式细胞术分析,感染LV5或LV5/Cdc25C后DC2.4细胞表面因子CD11C和CD54的表达均升高,以LV/Cdc25C DC2.4组最高,差异有统计学意义(P<0.05)。结论成功制备肝癌相关抗原Cdc25C过表达树突状细胞疫苗。
