摘要
为了系统了解斯氏副柔线虫的转录组特征并构建其转录组图谱,以不同发育阶段的斯氏副柔线虫为研究对象,应用Illumina Nextseq 500高通量测序技术平台对其进行转录组测序及生物信息学分析,结果每个样本均得到3GB的原始数据,经质量控制和拼接组装后,共获得128 454个Unigene,序列片段的平均长度与N50值分别为634和1 037bp;将Unigenes与COG,Nr,Trembl等数据库进行比对,结果表明有35 324个Unigene比对到COG数据库,根据蛋白种类大致分为25类;通过GO功能分类,5 227个Unigene映射到GO不同的功能节点上;通过KEGG pathways分析,共有13 424个Unigene参与了8 155个代谢通路。该研究结果为斯氏副柔线虫病的基础理论、诊断方法及防治研究奠定基础。
To determine the transcriptome of Parabronema skrjabini in camle and construct the transcriptome map, the transcriptome of the different stages of Parabronema skrjabini was analyzed using Illumina Nextseq 500-high throughput RNA sequencing. The results showed that each sample received more than 3 GB raw data. 128 454 Unigenes were obtained by qulity control and de novo assembly method,and the mean length and N50 length were 634 and 1 037 bp,respectively. All of Unigenes were annotated using the BLAST search against COG,Nr,Trernbl databases. The results analysis indicated 35 324 Unigenes were annotated in COG database and broadly divided in- to 25 classes according to the function. A total of 5 227 Unigenes were mapped into different GO terms. By KEGG database as a reference, 13 424 Unigenes were assigned to 8 155 KEGG path- ways. This study provides a foundation and reference for further investigation of fundamental theory,diagnosis methods and prevention on parabronemosis.
