检测条件对血液透析用水微生物监测结果的影响 被引量:4

Influence of testing conditions on microbial monitoring results in hemodialysis water

摘要 目的探讨培养基、标本接种时间、接种方式、培养温度、培养时间等因素对透析用水微生物监测结果的影响。方法采集某三甲医院血液透析用水标本360份,分别采用倾注法、涂布法于采样后4 h、8 h接种TGEA、R2A及普通营养琼脂,分别于20℃、30℃、35℃培养48~168 h,观察菌落生长情况并计数。结果三种培养基之间细菌培养结果差异无统计学意义;涂布法接种后透析用水细菌总数明显高于倾注法;采样8 h后接种,普通营养琼脂细菌培养结果明显高于采样4 h后接种;随着培养时间的延长,细菌总数增加;20℃及30℃培养条件下,透析用水细菌培养结果差异无统计学意义;35℃培养后细菌总数低于20℃和30℃。结论血液透析用水的微生物学监测宜于采样后4 h内使用涂布法接种于TGEA、R2A或普通营养琼脂,20℃或30℃培养168 h,有助于提高血液透析用水微生物学监测的准确性。 Objective To explore the influence of culture medium,inoculability time,inoculation type, culture temperature and culture time on the hemodialysis water tested by microbial monitoring. Methods 360 samples in a third - class - A hospital were collected. 8 h or 4 h later, the samples were inoculated by TGEA, R2A and Common nutrient agar with pouring method and coating method and were put into the conditions under the temperature of 20,30, and 35℃ respectively for 48 - 168 h. Then colony growth was observed was and counted. Results There was no significant difference in the three culture media of bacterial culture. The total amount of bacteria inoculated with coating method in the dialysis water was higher than the amount of pouring method significantly. In common nutrient agar, samples inoculated after sampling 8 h were higher than those inoculated after sampling 4 h. With the extension of culture time, the total number of bacteria of hemodialysis water had increased. There was no significant difference in the results of bacterial culture of hemodialysis water in the conditions under the temperature of 20℃ and 30℃. The total amount of bacteria in 35℃was lower than those in 20℃and 30℃. Conelusion TGEA, R2A and Common nutrient agar can be used for hemodialysis water monitoring, and the samples should be inoculated within 4 hours at 20℃ or 30℃ for 168 h . It is helpful to improve the accuracy for the monitoring results of hemodialysis water.
