摘要 目的研究宫颈癌组织中lncRNA的表达变化情况,为探讨lncRNA参与宫颈癌发生与发展提供分子依据。方法取20例宫颈癌患者的宫颈癌组织和20例正常宫颈组织,应用高通量lncRNA芯片技术分别检测两组中lncRNA表达谱的变化情况,并应用qRT-PCR技术对结果加以验证。结果与正常宫颈组织相比,宫颈癌组织中共有30586条lncRNA和13982条mRNA存在差异性表达,通过GO聚类分析表明,上调mRNA和下调mRNA分别参与多种不同的生物过程。通过进一步分析发现变化差异性最明显的10个lncRNA可能通过影响相应的mRNA来发挥其生物学功能。qRT-PCR结果与芯片检测结果一致,提示4个lncRNA(BC008359,RP11-521B24.5,BRE-AS1,和RP11-229P13.15)在宫颈癌组织中发挥重要作用。结论本实验阐述了宫颈癌组织中lncRNA的变化情况,为探讨lncRNA参与宫颈癌发生与发展以及治疗提供新的分子依据。 Objective To inves:igate the changes of lncRNA in cervical cancer tissues, and provide molecular basis for investigating the occurrence and development of cervical cancer related with lncRNA. Methods 20 cervical cancer patients and 20 normal cervical tissues were selected. A Dplicated with high-through put microarray lneRNA ,we tested the variation of lncRNA expression profiles of normal cervical tissue s and cervical cancer tissues. The result was verified by real-time quantitative RT- PCR. Results Compared with normal cervical tissues, a total of 30586 lncRNA and 13982 mRNA were identified as differentially expressed in cervical cancer tissues. GO cluster analysis showed that up-regulated mRNAs and down-regulated mRNAs were associated with various biological processes respectively. The lO lneRNA showed the most obvious difference analyzed to influence mRNA corresponding to exert its biological function. Quantitative PCR results were consistent with the microarray results suggested four lncRNA ( BC008359, RP11-521 B24.5, BRE-AS1, and RP11-229P13.15 ) played important roles in cervical cancer. Conclusion The changes of lncRNA in cervical cancer, and provide molecular basis for investigating the occurrence and development and the therapy of cervical cancer related with lncRNA.
