药物治疗后肾癌EMT相关基因表达谱分析及对预后的影响

Analysis of EMT-related Gene Expression Profile and Prognosis of Renal Cell Carcinoma after Drug Treatment

目的通过对相关数据库数据分析,探究EMT相关调节因子的异常表达与肾癌的相关性。方法对NCBI中GEO数据库中的若干数据集数据进行表达量分析、生存分析和相关性分析。结果 1研究分析表明,Snai1等EMT相关调节因子在肾癌中存在明显的高表达,而E-cadherin(CDH1)等则表现为明显的低表达。2Twist1等的高表达会导致肾癌患者的总生存率和无疾病生存率明显降低(P=0.0186;P<0.0015)。3VIM和SIP1在药物治疗后的肾癌患者中有明显的高表达情况(VIM,P<0.001;SIP1,P=0.0021),而CDH1和TJP1在药物治疗后的肾癌患者中则表现为低表达(CDH1,P<0.0002;OCLN,P<0.0001)。4VIM和SIP1在肾癌患者中的表达情况呈正相关(P<0.0001,γ=0.573);而其与OCLN呈负相关(P=0.0017,γ=-0.3316)。结论通过对EMT指标在肾癌患者中的表达相关性分析表明,EMT调节因子的异常表达与肾癌患者疾病的发展存在一定的相关性,并且对其药物耐受和预后都有一定的影响。

Objective To study relevant database data analysis,and explore the correlation of the abnormal expression of EMT related factors and renal cell carcinoma. Methods Based on some data sets of GEO in the NCBI database, to perform express analysis, survival analysis and correlation analysis. Results （1）The results showed that Snaill and other EMT-related regulatory factors in renal cell carcinoma patients were significantly higher expression, while the E-cadherin （CDH1）and so the performance was significantly lower expression. （2）Twistl and other high expression of renal cancer patients will lead to the overall survival and disease-free survival was significantly reduced（ P = 0. 0186 ;P 〈 0.0015 ）. （3）VIM and SIP1 were significantly higher in patients with renal cancer after drug treatment（ VIM ,P 〈 0. 001 ; SIP1 ,P = 0.0021 ） ,while CDH1 and TJP1 showed low expression in renal cancer patients after drug treatment （ CDH1 ,P 〈 0. 0002 ; OCLN, P 〈 0. 0001 ）. （4）There was a positive correlation between the expression of VIM and SIP1 in renal cell carcinoma（P 〈0. 0001 ,γ=0. 573 ）and negative correlation with OCLN（P = 0. 0017,γ= -0.3316）. Conclusion The correlation analysis between the EMT index and the expression of RCC in RCC patients showed that the abnormal expression of EMT regulatory factor is correlated with the development of RCC and has certain influence on drug tolerance and prognosis.
