摘要
目的:观察自拟麻枳化浊方对糖尿病胃轻瘫(DGP)大鼠胃窦Cajal间质细胞(ICC)分布数量的影响。方法:将40只健康雄性Wistar大鼠随机分为正常组、模型组、中药组、西药组、联合组,模型组和正常组分别给予同等体积的生理盐水灌胃,中药组给予麻枳化浊方、西药组给予西沙必利、联合组给予西沙必利加麻枳化浊方进行干预治疗;在光镜下观察大鼠肠道组织的病理改变,应用免疫组化染色法检测各组大鼠胃窦组织中c-kit抗体阳性ICC细胞的表达及分布特点,并使用FR-980复日生物电泳图像分析系统测定大鼠胃窦c-kit抗体免疫反应阳性产物的平均光密度(A)值。结果:中药组、西药组及联合组大鼠小肠推进率较模型组均明显增加(P<0.05);模型组大鼠胃窦部c-kit抗体平均光密度值减少,表达减弱(P<0.05),中药组、西药组与联合组尚可见着色较深的c-kit阳性细胞,其平均光密度值较模型组升高(P<0.05),且中药组及联合组平均光密度值均明显高于西药组(P<0.05)。结论:麻枳化浊方能够降低DGP大鼠血糖水平,提高其小肠推进率,可能为其治疗DGP的作用机制之一;麻枳化浊方能够增加DGP大鼠胃窦部c-kit阳性细胞的平均光密度,可能通过调控其胃窦部Cajal ICC的数量,促进大鼠的胃排空。
Objective:To observe the effects of Mazhi Huazhuo decoction on the number distributions of interstitial cells of Cajal of rats with diabetic gastroparesis(DGP).Methods:Total 40 healthy male Wistar rats were randomly divided into normal group,model group,traditional Chinese medicine group,western medicine group and combined group.The treatment groups were given Mazhi Huazhuo decoction,cisapride and Mazhi Huazhuo decoction combined with cisapride.The model group and normal group were gavaged with same volume of saline.Pathological changes of intestinal tissue were observed under light microscope,immunohistochemical staining was used to detect the expression and distribution of c-kit positive ICC cells in gastric antrum of rats.The average optical density values of immunoreactive products of c-kit antibody in gastic antrum of rats were measured by FR-980 complex day electrophoresis image analysis system.Results:The intestinal propulsive rates of traditional Chinese medicine group,western medicine group and combined group were significantly increased compared with the model group(P〈0.05).The average optical density values of c-kit antibody and expressions in the gastric antrum of rats in model group decreased(P〈0.05).Dark-stained c-kit positive cells were seen in Chinese medicine group,western medicine group and combined group,the average optical density values higher than the model group(P〈0.05)and the average optical density values of the traditional Chinese medicine group and combined treatment group were significantly higher than those in the western medicine group(P〈0.05).The average optical density between traditional Chinese medicine group and combined group had no significant difference(P〉0.05),more than western medicine group(P〈0.05).Conclusion:Mazhi Huazhuo decoction could reduce the blood glucose levels of DGP rats and improve the intestinal propulsive rate which might be one of the possible mechanisms for the treatment of DGP.Mazhi Huazhuo decoction could increase the average optical density of c-kit positive cells in gastric antrum of rats with DGP,probably by regulating the number of gastric antrum interstitial cells of Cajal(ICC)and promoting gastric emptying in the treatment of rats with DGP.
