电针对2型糖尿病大鼠糖代谢及血清SOD、MDA含量的影响 被引量:11

Effect of Electro-acupuncture on Oxidative Stress and Glucose Metabolism in Rats with Type 2 Diabetes

摘要 目的:观察电针胃脘下俞与后三里对2型糖尿病(T2DM)大鼠糖代谢和氧化应激的影响,探讨电针改善T2DM的可能作用机制。方法:雄性SD大鼠随机分为空白组、模型组、胃脘下俞组、后三里组和格列美脲组,每组8只。造模方法为高糖高脂饲料饲养联合小剂量链脲佐菌素(Streptozotocin,STZ)35 mg/kg一次性腹腔注射。电针(2 Hz、2 mA)干预,每次留针20 min,6次/周,共4周。测量大鼠体质量及空腹血糖;采用放射性免疫法检测大鼠血清中空腹胰岛素(FINS)水平、丙二醛(MDA)含量及超氧化物歧化酶(SOD)水平,并于干预前后进行口服糖耐量试验(OGTT);计算稳态模型胰岛素抵抗指数(HOMA-IR)及胰岛B细胞功能指数(HOMA-B)。结果:电针"胃脘下俞"及"后三里"能在1~2周左右显著降低模型动物血糖。通过电针干预均在一定程度上降低T2DM大鼠的空腹胰岛素水平、胰岛素抵抗指数,且胃脘下俞组最优(P<0.01)。与空白组比较,模型组大鼠的空腹血糖和血清MDA含量均升高,SOD含量均降低(P<0.05);与模型组比较,胃脘下俞组、后三里组SOD含量均升高(P<0.05),MDA含量均降低(P<0.05),而格列美脲组SOD、MDA含量与模型组相比,差异无统计学意义(P>0.05)。结论:电针胃脘下俞穴和后三里对T2DM大鼠均具有一定的降糖作用,并可能通过整体上改善其氧化应激状态来改善胰岛素抵抗,进而达到干预T2DM的作用。 Objective: To observe the effect of electro-acupuncture( EA) on oxidative stress and glucose metabolism in rats with type 2 diabetes( T2DM),so as to study its mechanism of improving T2 DM. Methods: 40 male SD rats were evenly randomized into control,model,"Weiwan Xiashu","Housanli "and Glimepiride groups,with 8 rats in each group. The T2 DM models were established by feeding high fat and sugar diet combined with intra-peritoneal injection of streptozotocin( 35mg/kg). Electro-acupuncture( 2 Hz,2 m A) was applied as the intervention,20 minutes every time,6 times per week,lasting for 4 weeks. Body quality and fasting blood glucose( FBG) were measured once a week during the intervention. Fasting serum insulin( FINS),serum malondialdehyde( MDA) and superoxide dismutase( SOD) levels were determined by using radioimmunoassay. Oral glucose tolerance test was measured before and after the intervention. In addition,homeostasis model assessment for insulin resistance( HOMA-IR) and modified B cell function index( MBCI) were calculated. Results: The fasting blood glucose was decreased to a certain extent with the application of electro-acupuncture at Weiwan Xiashu and Housanli. After 4 weeks,FINS and HOMA-IR of T2 DM rats were decreased,and the Weiwan Xiashu group was the best( P〈0. 01). Compared with those of the control group,FBG and MDA levels of model group were both increased. However,SOD activities were decreased( P〈0. 05). Compared with those of the model group,SOD activities of Weiwan Xiashu,Housanli groups were increased( P〈0. 05)and MDA levels were decreased( P〈0. 05). There was no statistical significance between the Glimepiride group and the model group( P〉0. 05). Conclusion: Electro-acupuncture at Weiwan Xiashu and Housanli can reduce fasting blood glucose and may normalize insulin sensitivity,ameliorate insulin resistance and hyperinsulinemia by improving the oxidative stress state as a whole.
