皖南红豆杉茎、叶多糖抑制Lewis肺癌小鼠肿瘤机制研究 被引量:1

Study on the Antitumor Mechanism in Lewis Lung Cancer Mice with the Stem and Leaf Polysaccharides in the South Taxus of Anhui Province

摘要 目的:研究皖南红豆杉茎、叶多糖抑制Lewis肺癌小鼠肿瘤及其机制。方法:以Lewis肺癌小鼠为模型,观察茎、叶多糖对瘤重及瘤组织中的血管内皮细胞生长因子(VEGF)、碱性成纤维细胞生长因子(bFGF)、血管抑素(Angiostatin)、内皮抑素(Endostatin)、微血管密度(MVD)的影响。结果:茎、叶多糖可明显抑制移植瘤生长,降低VEGF、bFGF表达、MVD,增加Angiostatin、Endostatin表达,其中YX leaf多糖中、低剂量组抑瘤作用,降低VEGF、bFGF表达、MVD,增加Endostatin表达上均优于TP stem多糖相应组别(P<0.05);YX leaf多糖高、低剂量组在提高Angiostatin表达上优于TP stem多糖相应组别(P<0.05)。结论:皖南红豆杉茎、叶多糖抑制肺癌机制可能与降低VEGF、bFGF的水平,降低MVD,提高Angiostatin、Endostatin的水平有关,且与吉非替尼联用后药效增强。 Objective: To observe the antitumor mechanism in lewis lung cancer mice with the stem and leaf polysaccharides in the south taxus of Anhui province on tumor in Lewis lung cancer mice. Methods: Taken the lewis lung cancer mice as the model, the antitumor effect and expressions of VEGF, b FGF, angiostatin, endostatin and MVD in the tumor tissue were evaluated. Results: The stem and leaf polysaccharides inhibited significantly the growth of the transplanted tumor, reduced the expression of VEGF 、 b FGF and MVD and improved the expression of Angiostatin and Endostatin, in which the medium and low doses YX leaf polysaccharides group had higher effect of lowering the expression of VEGF, b FGF and MVD and increasing the expression of endostatin than that of corresponding TP stem polysaccharides group ( P 〈 0. 05) , and the high and low doses YX leaf polysaccharides group had higher effect of increasing the expression of angiostatin than that of corresponding TP stem polysaccharides group ( P 〈 0. 05 ) . Conclusion: The antitumor mechanism of the stem and leaf polysaccharides in the south taxus of Anhui province might be achieved through lowering the angiogenesis factor level of VEGF, b FGF, abating the MVD and enhancing the angiostatin and endostatin level. The gefitinib plus the stem or leaf polysaccha- rides can enhance the effect.
