水稻苗期耐盐性基因SST分子标记的筛选与应用 被引量:2

摘要 以水稻耐盐突变体sst为对照,对67个水稻亲本及突变体sst与水稻品种华占的F2群体进行耐盐性筛选,并结合分子标记结果,分析水稻苗期耐盐性基因SST分子标记ID27093、ID27101、ID27118、INDEL1和INDEL2的特异性和准确性.结果表明,分子标记ID27093、ID27101、ID27118和INDEL1的扩增多态性较低;而分子标记INDEL2的扩增多态性较高,为共显性标记,具有较高的特异性,能区分耐盐供体sst与85.07%的供试亲本,且筛选准确率高.因此,分子标记INDEL2标记可以直接用于耐盐性基因SST分子标记辅助育种. Molecular markers,INDEL1 and INDEL2,which are linked to salt tolerance gene SST of rice seedling,were designed.The specificity and reliability of 5 markers,including ID27093,ID27101,ID27118,INDEL1,and INDEL2,were analyzed through screening 67 rice varieties and a F2 population from a cross between mutant sst and a rice restorer line Huazhan for salt tolerance,and mutant sst was used as a salt tolerance control. The results showed that all markers except INDEL2 demonstrated low polymorphism. INDEL2 turned out to be a co-dominant marker with high specificity,and it was able to distinguish from 85.07% of the tested parents and mutant sst with high screening accuracy. Therefore,marker INDEL2 could be directly used as a molecular marker in breeding for rice seedling salt tolerance gene SST.
