蜂胶可吸收成分及其特征单体抑制心肌细胞凋亡实验研究

摘要 观察蜂胶可吸收成分及其特征黄酮单体对阿霉素诱导大鼠心肌细胞凋亡模型的保护作用。从大鼠灌胃蜂胶提取物及其特征单体白杨素、高良姜素30 min后,腹主动脉取血,离心制备含蜂胶及特征单体的血浆。提取SD大鼠乳鼠心肌细胞并培养,采用阿霉素制备心肌细胞凋亡模型;将制备的蜂胶及白杨素、高良姜素的血浆与心肌细胞混合,使用缺口末端标记法(TUNEL法)进行心肌细胞凋亡检测。实验结果表明,蜂胶提取物、白杨素、高良姜素组可吸收成分心肌组织细胞凋亡率与模型组比较显著降低,差异有统计学意义(P<0.05);蜂胶提取物组效果较好,蜂胶可吸收成分具有心肌细胞保护作用,减轻阿霉素诱导的心肌损伤。 To observe the effect on protection on cardiomyocyte apoptosis of mice induced by adriamycin cells by ab-sorbable compounds of propolis and its characteristic flavonoid compounds. The rats were perfused with extract of propolis and its characteristic flavonoids: Chrysin and galangin. After 30 min, blood were collected from abdominal aorta and prepared different centrifuged plasma : extract of propolis, Chrysin and galangin. Extracted ventricular cells from suckling mice and cultivated, then prepared cell model of extract of propolis by adriamycin. Mixed differ-ent plasma: normal, extract of propolis, Chrysin and galangin with cells, then used Terminal dUTP Nick End La-beling (TUNEL) method to do detection of cardiomyocyte apoptosis. Result shows that group of extract of propolis, Chrysin and galangin can more obviously decrease the rate of cardiomyocyte apoptosis than that of model group, with statistical significance(P 〈0. 05) ; the group of extract of propolis shows the best effect. Adsorbable element of propolis can significantly protect ventricular cells,and alleviate damage of ventricular cells caused by adriamycin.
