摘要 目的探讨Synaptotagmin1(Syt1)对小鼠卵母细胞减数分裂过程和皮质反应的影响。方法免疫印迹检测Syt1在小鼠卵母细胞的表达;免疫荧光分析Syt1在卵母细胞中定位以及对减数分裂过程的影响;活细胞实时拍摄系统观察皮质反应以及纺锤体、染色体在降调Syt1表达的动态变化。结果 (1)Syt1和小鼠卵母细胞皮质颗粒完全共定位;(2)降调Syt1表达,小鼠卵母细胞减数分裂进程受到阻碍;(3)活细胞实时拍摄系统证实了降调Syt1会影响中后期转换、第一极体释放以及进一步的皮质反应。结论 Syt1参与调控卵母细胞的减数分裂的成熟过程和皮质反应的发生。 Objective:To identify the effect of Synaptotagmin 1 (Syt1) on meiosis and cortical reaction in mouse oocyte.Methods:Syt1 expression was determined by immunoblotting analysis.The localization of Syt1 in oocytes and the effect on meiosis processes were analyzed by immunofluorescence.Cortical reaction and the dynamics of microtubule and chromosome were filmed with Perkin Elmer precisely Ulta VIEW VOX Confocal Imaging System.Results:Syt1 expression was localized at the same position as cortical granules(CGs) in mouse oocyte.Down-regulation of Syt1 expression interrupted the meiotic process in mouse oocyte.The results of Perkin Elmer precisely Ulta VIEW VOX Confocal Imaging System confirmed that down-regulation of Syt1 affected MI and MII conversion,the first polar body release and further cortical response.Conclusions:Syt1 is involved in mouse oocyte meiotic maturation process and cortical reaction.
