摘要
目的:评价加味逍遥散对兔去势干眼症的疗效,为临床治疗提供依据。方法:将40只日本成年雌性大耳兔手术摘除双侧卵巢,制作成干眼症模型40例(80眼),随机均分为5组:模型组(A)、对照组(B)、加味逍遥散低剂量组(C)、加味逍遥散中剂量组(D)和加味逍遥散高剂量组(E),每组8只。对5组实验兔进行去势术建立动物模型,对照组口服黄体酮胶囊,C、D、E组分别给于5、10、20倍浓度加味逍遥散连续灌胃45d,测定给药后兔去势干眼症经加味逍遥散治疗前后泪腺组织中炎症因子IL-1β、TNF-α、TGF-β水平、基础泪液分泌试验(schirmer I test,SIT)、泪膜破裂时间(break-up time,BUT)及角膜荧光素染色(corneal fluorescent staining,Fl)情况的改变,治疗前后及5组间均进行统计学分析。结果:C、D、E组SIT及Fl试验测量值明显高于A组(P<0.05)和B组(P<0.05),BUT试验B组和C组没有差别(P>0.05),在泪腺组织中,IL-1β、TNF-α阳性表达的细胞数均低于B组,而TGF-β1阳性表达的细胞数均明显高于B组(P<0.01)。以上检测结果 E组均优于C、D组(P<0.05)。结论:加味逍遥散可显著改善兔去势干眼症的实验室检查指标,其作用机制可能与加味逍遥散具有类似于雌激素作用有关,可起到促雌激素效应,从而调节泪腺局部炎症反应有关,具有重要的临床应用价值,为围绝经期干眼症临床治疗提供实验参考。
Objective: To observe influences of the modified Xiaoyao Powder on dry eye in castrated rabbits provide the basis data for clinical treatment. Methods: Surgical removal of bilateral ovaries for forty female rabbits to made dry eye model of 40 cases( 80 eyes),randomly divided on average into 5 groups: model group( A),control group( B),modified Xiaoyao Powder middle dose group( C),modified Xiaoyao Powder low dose group( D),modified Xiaoyao Powder high dose group( E),each group of 8 rabbits. For 5 groups of experimental rabbits castration establishment of animal model,the control muscle per os of progesterone,C、D、E group respectively for5,10,20 times the concentration of continuous lavage 45 day,determination of rabbits castration dry eye after the treatment by the modified Xiaoyao Powder before and after the treatment of lacrimal gland tissue inflammation of IL- 1β,TNF- α and TGF- β,basic tear secretion test( Schirmer I test,SIT),it appears rupture time( break- up time,BUT) and corneal fluorescein staining( corneal fluorescent staining,Fl) the change of the situation,the statistical analysis and 5 groups before and after treatment. Result: The SIT and Fl values for Groups C,D,E were significantly higher than those in group A( P 〈 0. 05) and group B( P 〈 0. 05),BUT experimental for group B and group C there was no difference( P 〉 0. 05),in lacrimal gland organization,IL- 1β,TNF- α positive expression cells were lower than group B,and positive expression of TGF- β cell count were significantly higher than that of group B( P〈 0. 01). E groups are better than C,D groups in the above test results( P 〈 0. 05). Conclusion: Modified Xiaoyao Powder can sig-nificantly improve the rabbits castration laboratory examination indexes of dry eye,its mechanism may be related to modified Xiaoyao Powder has a role similar to estrogen,can rise to promote the effect of estrogen,which regulates lachrymal local inflammatory response,has important clinical value,to provide experimental reference for dry eye for perimenopausal clinical treatment.
