摘要
利用荧光光谱和圆二色谱研究了细胞色素 c氧化酶 Cu A 结构域蛋白的稳定性。结果表明 ,随着溶液p H值的升高 ,Cu A 结构域蛋白的稳定性降低。随着温度的升高 ,Cu A结构域蛋白的二级结构遭到破坏。脱金属形式蛋白的热稳定性减小 ,表明双核金属铜中心的存在对 Cu A
The stability of Cu A domain of cytochrome c oxidase was investigated by fluorescence spectra and circular dichroism spectra.The results showed that the stability of Cu A domain decreased with the increasing of pH,and its secondary structure was destroyed with the increase of temperature.The thermal stability of apo-Cu A domain is higher than that of Cu A domain, which suggested that the binuclear metal center [Cu A +1.5-Cu A +1.5] also plays an important role on the structural stability of the protein.
