摘要
用金属指示剂铬黑 T(EBT)为共振光散射探针 ,对敦煌壁画含不同颜料的胶结材料中的蛋白质含量进行了定量测定。在 p H=4 .10的 Britton- Robinson缓冲溶液条件下 ,在波长 λ=375 nm处 ,以牛血清白蛋白 (BSA)为标准样品绘制校准曲线 ,测定含不同颜料的胶结材料中蛋白质结果分别为 :黄色颜料8.715 1μg/ mg;红色颜料 6 .30 71μg/ mg;黑色颜料 6 .16 5 5 μg/ mg;绿色颜料 2 .2 2 6 5 μg/ mg和白色颜料1.10 6 6 μg/ mg。对 BSA的检出限为 5 5 μg/ L,线性范围 0— 13mg/ L。该方法简单、快速、灵敏度高 ,平行测定的相对标准偏差 (RSD)为 2 .7% ,蛋白质的加标回收率为 96 .7%— 10 6 .6 %。
A novel method for the determination of proteins in the binding media of wall paintings was established based on the interaction between Eriochrome Black T(EBT) and proteins,which results in resonance light scattering(RLS) in the Britton Robinson buffer at pH 4.10.Bovine serum albumin (BSA) was employed as the original analogue of proteins in wall paintings and RLS intensity at 375nm is proportional to the concentration of proteins in the range 0—13mg/L.8.7151μg, 6.3071μg, 6.1655μg,2.2265μg and 1.1066μg proteins were detected for 1mg yellow,red,black,green and white pigments of wall paintings at Dunhuang,respectively.The method is simple,rapid and sensitive,the detection limit is 55μg/L and the relative standard deviation is 2.7%.The recovery is between 96.7% and 106.6%.
