摘要
石油醚∶丙酮 (7∶ 3)混合液作提取剂 ,薄层层析 -分光光度法测定了南瓜中 β-胡萝卜素的含量 ,对该法的精密度、准确性进行了探讨。在 β-胡萝卜素的最大吸收波长 4 5 0 nm下 ,回归方程为 A=0 .0 2 4 0 5 6 C-0 .0 0 2 4 6 4,r=0 .998。测定结果的相对标准偏差在 2 .0 1%— 4 .5 2 %之间 ,回收率在 94 .5 0 %— 97.2 0 %之间 ,检出限为 0 .5
Thin layer chromatography-spectrophotometry was used to determine the content of β-carotene in pumpkin extracted with the mixture of petroleum ether acetone (7∶3,V∶V).The precision,accuracy and stability of the method were studied.The standard linear equation was A=0.024056C-0.002464,r=0.998 with the relative standard deviation of 2.01%—4.52%.The recovery of β-carotene is in the range of 94.50%—97.20% with the detection limit of 0.5μg.
