摘要
目的探讨骨髓增生异常综合征(MDS)患者外周血单个核细胞(PBMC)中视网膜母细胞瘤蛋白结合锌指结构基因1(RIZ1)的表达及其临床意义。方法选取2012年7月至2015年7月于新乡医学院第一附属医院收治的MDS患者86例为研究对象(MDS组),并选取同期体检健康者90例作为对照组,采用实时荧光定量聚合酶链反应和Western blot法检测PBMC中RIZ1蛋白及其mRNA表达水平,并分析RIZ1表达与MDS患者临床病理特征的关系。结果对照组和MDS组受试者PBMC中RIZ1 mRNA表达水平分别为1.73±0.41和0.47±0.18,MDS组患者PBMC中RIZ1 mRNA表达水平显著低于对照组(P<0.05)。对照组和MDS组受试者PBMC中RIZ1蛋白表达水平分别为1.32±0.36和0.35±0.22,MDS组患者PBMC中RIZ1蛋白表达水平显著低于对照组(P<0.05)。RIZ1蛋白表达与MDS患者的年龄、性别、白细胞计数、血红蛋白水平及血小板计数无关(P>0.05),而与骨髓原始细胞比例、世界卫生组织(WHO)分型及染色体核型有关(P<0.05)。结论 RIZ1在MDS患者PBMC中呈低水平表达,并与患者的骨髓原始细胞比例、WHO分型及染色体核型显著相关,这为MDS的早期诊断和靶向治疗提供了新的思路。
Objective To investigate the clinical significance of the expression of retinoblastoma protein-interacting zinc finger gene 1( RIZ1) in peripheral blood mononuclear cell( PBMC) of patients with myelodysplastic syndrome( MDS).Methods Eighty-six patients with MDS in the First Affiliated Hospital of Xinxiang Medical University from July 2012 to July2015 were selected as MDS group,and ninety healthy subjects were selected as control group. The expressions of RIZ1 protein and mRNA in PBMC were detected by real time fluorescence quantitative polymerase chain reaction and Western blot,and the relationship between RIZ1 expression and clinicopathological features of MDS patients was analyzed. Results The expression of RIZ1 mRNA in PBMC in the control group and MDS group was 1. 73 ± 0. 41 and 0. 47 ± 0. 18,respectively; the expression of RIZ1 mRNA in PBMC in the MDS group was significantly lower than that in the control group( P 〈 0. 05). The expression of RIZ1 protein in PBMC in the control group and MDS group was 1. 32 ± 0. 36 and 0. 35 ± 0. 22,respectively; the expression of RIZ1 protein in PBMC in the MDS group was significantly lower than that in the control group( P 〈 0. 05). The expression of RIZ1 protein was not associated with age,sex,white blood cell count,hemoglobin level and platelet count in patients with MDS( P 〈 0. 05); but it was significantly correlated to the proportion of bone marrow progenitor cells,world health organization( WHO) typing and chromosome karyotype( P 〈 0. 05). Conclusion The expression of RIZ1 decreases in PBMC of MDS patients,and the expression of RIZ1 is significantly correlated to the proportion of bone marrow progenitor cells,WHO typing and chromosome karyotype; which provides a new way for early diagnosis and targeted therapy of MDS.
