黑曲霉菌种的筛选及培养条件的优化 被引量:4

摘要 本文培养黑曲霉,选取产酶活力最好的黑曲霉,通过对黑曲霉培养的主要条件,温度、p H、装液量、接种量、碳源以及氮源等条件的优化,提高黑曲霉产果胶酶活力。菌种M1、M2、M3产果胶酶、木聚糖酶和纤维素酶活力比KC1、KC2、KC3高。其中M1产果胶酶活力最高,产木聚糖活力较高。通过对M1产果胶酶、木聚糖酶、纤维素酶活力的比较,确定了最佳培养温度为30℃,pH4.0,接种量为5%,250mL锥形瓶装液量为50mL时,酶活力达到最大。通过对培养基碳源、氮源的优化,确定了最佳碳源为麸皮,最佳氮源为(NH_4)_2SO_4。 In this paper,fristly by selecting the best enzyme activity of Aspergillus niger,and changing the culture conditions of Aspergillus niger( p H,optimal temperature,liquid volume,inoculation amount,carbon source and nitrogen source),the activity of enzyme produced by Aspergillus niger was improved. The activity of pectinase,xylanase and cellulase produced by M1,M2,M3 is higher than that of KC1,KC2,KC3. The activity of pectinase produced by M1 is the highest,and xylanase with high activity. The results showed that the best fermentation temperature was 30℃,pH = 4. 0,the best inoculation volume was 5%,and loaded liquid volume was 50 mL in conical flask with 250 mL. The best carbon source was bran,the best nitrogen source was(NH4)2SO4.
