摘要
目的探讨DJ-1在胰腺癌中的表达及与患者预后的关系。方法选取64例胰腺癌患者作为胰腺癌组,选取门诊健康体检者作为对照组,胰腺癌组根据DJ-1的表达水平进行亚分组(高DJ-1组和低DJ-1组)。采用酶联免疫吸附试验(ELISA)检测两组的血清DJ-1和CA19-9水平,统计不同TNM分期的胰腺癌患者的血清DJ-1和CA19-9水平,分析DJ-1与CA19-9水平的相关性及DJ-1水平与患者生存率的关系。结果胰腺癌组患者的血清DJ-1水平高于对照组,差异有统计学意义(P﹤0.05)。随着TNM分期的增加,胰腺癌患者的血清DJ-1和CA19-9水平随之升高(P﹤0.05)。胰腺癌患者血清DJ-1与CA19-9水平呈显著正相关(P﹤0.01)。高DJ-1组较低DJ-1组存在较高比例的肝脏及淋巴结转移,且肿瘤大小及TNM分期较低DJ-1组高(P﹤0.05)。术后14周后,高DJ-1组的生存率迅速降低,两组患者的生存率比较,差异有统计学意义(P﹤0.05)。结论 DJ-1在胰腺癌患者的血清中高表达,DJ-1高表达的胰腺癌患者TNM分期较高,生存率较低,提示DJ-1水平可能为胰腺癌患者预后的生物标志物之一。
Objective To explore the expression of DJ-1 in pancreatic cancer and the association with patient's prog-nosis. Method 64 cases of pancreatic cancer patients were included in the research as study group, and at the same time matched healthy volunteers were enrolled as control group, while the study group was further subdivided according to the expression of DJ-1 (as subgroups of high or low DJ-1 expression). Serum CA19-9 and DJ-1 levels in the two groups were detected by ELISA assay and statistically analyzed in patients with different TNM stages, the association between DJ-1 level and CA19-9, as well as DJ-1 and survival were investigated. Result Pancreatic cancer patients had significantly higher serum DJ-1 level than normal volunteers (P〈0.05). Cancer patients with higher TNM stages had elevated serum DJ-1 and CA19-9 levels (P〈0.05). There was remarkable positive correlation between DJ-1 and CA19-9 level in pancreat-ic cancer patients(P〈0.01). While in subgroups, higher DJ-1 expression were correlated with more liver metastasis and lymph node metastasis, larger tumor size, and higher TNM stages compared with those with lower DJ-1 expression(P〈0.05). In 14 weeks after surgery, it was observed that patients with higher DJ-1 level had a significant fall in survival rate in comparison with patients with lower DJ-1 level, and the two subgroups differed significantly in respect of survival rate (P〈0.05). Conclusion The serum level of DJ-1 is high in pancreatic cancer patients, and those with higher DJ-1 expres-sion are more likely to have advanced TNM stages and significantly lower survival rate, suggesting that DJ-1 may be a biomarker in predicting the prognosis of pancreatic cancer patients.
