摘要 目的:以白芨假鳞茎为培养材料,进行愈伤组织诱导和抗褐化研究.方法:使用兰科常用培养基MS,B5,KC分别作为基础培养基,应用正交试验方法设计生长调节剂配比.结果:当基础培养基为MS培养基时,并且6-BA1.5mg/L+2,4-D3.0mg/L时,愈伤组织诱导率最高,达73.3%.另外,添加活性炭可防止外植体褐化:活性炭含量为1.5g/L时,褐化率为44%,愈伤组织诱导效果最好;活性炭含量为3.0g/L时,褐化率最低为11%,愈伤组织诱导效果不好.结论:白芨愈伤组织诱导的最佳培养基为MS+6-BA1.5 mg/L+2,4-D3.0mg/L+AC2 g/L. Objective:In order to study the anti-browning and induction of Bletilla striata callus by tak- ing the false bulbs of Bletilla Striata as tissue culture material. Methods:The medium of MS, B5 and KC were selected as the basic culture medium of orchidaceae respectively. The ratio of growth regula- tors was adjusted via orthogonal tests. Results:The induction ratio was the maximum of 73.3% when MS medium was used as basic culture medium with 6-BA 1.5 mg/L + NAA 0mg/L + 2,4-D 3.0 mg/L. Additionally, addition the activated carbon into the basic culture medium could prevent Bletilla striata browning. The browning rates were 44% and 11% when the contents of activated carbon were 1.5 g/ L and 3.0g/L. The induction efficacy of Bletilla striata callus was the highest and least for both cont- ensts respectively. Conclusion:The optimal medium for induction of callus was MS + 1.5mg/L of 6-BA + 0mg/L of NAA +3.0 mg/L of 2,4-D +2g/L of AC.
