摘要
目的:通过对23d龄大鼠行颈背部皮下注射脱氢表雄酮(DHEA)建立多囊卵巢(PCO)大鼠高雄激素血症模型,为相关实验提供模型参考依据和临床治疗提供诊治思路。方法:将40只23d龄大鼠随机分成两组,即空白组、模型组,每组20只,模型组大鼠每天定期行颈背部皮下注射DHEA 6mg/(100g体质量·d)+注射用大豆油0.2m L;空白组同时间行颈背部皮下注射注射用大豆油0.2m L,两组均连续注射30d。实验期间监测造模大鼠的动情周期;实验结束后测量大鼠体质量、卵巢重量及观察卵巢形态学改变;用酶联免疫法测定血清相关激素,比较两组总睾酮(TT)、游离睾酮(FT)、促黄体生成素(LH)、雌二醇(E_2)、促卵泡生成激素(FSH)的水平值。结果:模型组大鼠阴道脱落细胞均为角质细胞,未见明显动情周期。模型组卵巢重量显著高于空白组,组间比较,差异有统计学意义(P<0.05);形态学检查结果显示模型组卵巢卵泡明显多于空白组,成多囊样发育,见多个明显囊状扩张卵泡。模型组TT、FT、E_2、LH水平值明显高于空白组,组间比较,差异均有统计学意义(P<0.05)。结论:DHEA可升高大鼠TT、FT、E_2、LH水平值,并使卵巢呈多囊样改变,成功建立了多囊卵巢大鼠高雄激素血症模型。该方法简单易行,是较为理想的造模方法,值得推广应用。
Objective :To establish a hyperandrogenism model in rats with polycystic ovary syndrome (PCOS)by subcutaneously injecting 23 -day -old rats with dehydroepiandrosterone(DHEA) in the neck and back, and to pro- vide a model reference for related experiments and a diagnostic and therapeutic idea for clinical treatment. Methods : Forty 23 - day - old rats were randomly and equally divided into blank group and model group. The rats in model group were subcutaneously injected with DHEA (6mg/100g body weight/day)and soybean oil (for injection, 0.2mL) in the neck and back every day. Meanwhile,the rats in blank group were subcutaneously injected with soy- bean oil(tbr injection,0.2mL) in the neck and back at the same time. Injection was given for 30 days in both groups. The estrous cycle of model rats was monitored during the experiment. At the end of experiment, the body weight and ovary weight were measured,and the morphological changes of the ovary were evaluated. ELISA was used to measure serum levels of total testosterone ( TY), free testosterone ( FT), luteinizing hormone ( LH ), estradiol ( E2 ), and follicle -stimulating hormone (FSH), which were then compared between the two groups. Resuhs:In the rats in model group, the shedding cells from the vagina were keratinocytes, without an obvious estrous cycle. The model group had a significantly higher ovary weight than the blank group ( P 〈 0.05 ) ; compared with the blank group, the model group had a significantly larger number of ovarian follicles, with polycystic development and many follicles with marked cystic dilatation. The model group had significantly higher TT, FT, E2, and LH levels than the blank group (P 〈 0. 05 ). Conclusion : DHEA can increase the serum levels of TT, FT, E2, and LH in rats and help to sue- eessfully establish a rat model of polycystic ovary and hyperandrogenism. The method is simple and ideal for model establishment and holds promise for application.
