乌鲁木齐地区维吾尔族、汉族哮喘儿童ADAM33 V4基因多态性与IgE水平的相关性研究 被引量:5

摘要 目的探讨乌鲁木齐地区维吾尔族、汉族支气管哮喘儿童ADAM33 V4基因多态性、IgE水平并进行相关性分析。方法选取同期3-15岁于乌鲁木齐地区生活的维吾尔族、汉族哮喘患儿197人(其中汉族111,维吾尔族86),同时设立对照组120人(汉族64,维吾尔族56),首先采用PCR聚合酶链反应对ADAM 33基因V4位点进行单核苷酸多态性(SNP)分型,并对其中部分进行基因检测验证。采用ELISA法检测IgE水平,进一步根据民族进行组内分层,进行V4位点SNP、IgE分析并讨论二者相关性。结果①ADAM33基因V4位点3种基因型分布差异有统计学意义(2=9.598,P=0.008);等位基因频率分布差异亦有统计学意义(2=12.500,P=0.000,OR=1.773,95%CI=1.289~2.438)。②与CC基因型比较,G等位基因的携带者(GG、CG+GG)基因型均能明显增加哮喘发生的危险(P=0.012、0.002,OR=2.469、2.406,95%CI=1.214~5.021,1.367~4.234)。③V4位点3种基因型频率分布在维吾尔族哮喘儿童中分布有统计学意义(2=6.998,P=0.030),汉族哮喘儿童中分布无统计学意义(2=4.994,P=0.082)。④哮喘组IgE水平高于对照组,差异有统计学意义(2=13.250,P=0.000)。④哮喘组组内雏吾尔族支气管哮喘儿童IgE水平高于汉族支气管哮喘儿童,差异有统计学意义(2=2.434,P=0.016);对照组组内维吾尔族、汉族儿童IgE水平比较无统计学差异(2=1.072,P=0.286)。⑤ADAM 33基因V4位点与IgE不相关(r=0.033,P=0.563)。结论①ADAM33基因V4位点与乌鲁木齐地区维吾尔族哮喘儿童发病相关,是该地区维吾尔族哮喘儿童的易感基因。②维吾尔族哮喘患儿IgE水平高于汉族族哮喘儿童。③ADAM 33基因V4位点不影响IgE水平。 Objective To investigate the Correlation of ADAM33 V4 gene polymorphism and IgE levels among Uygur and Han children with asthma in Urumqi.Methods 197 cases including 111 Han and 86 Uygur asthmatic children among 3 to 15 years old in Urumqi were selected,while control group was 120 cases including 64 Han and 56 Uygur normal children.At first the polymerase chain reaction and restriction fragment length polymorphism(PCR-RFLP) were used to analyze ADAM33 V4 gene single nucleotide polymorphism(SNP),in which part of samples were detected for genetic verification.ELISA method was used to measure the IgE levels,and correlation between IgE levels and ADAM33 V4 gene single nucleotide polymorphism was analyzed further according different nationality.Results The genotype frequencies of V4 Locus and allele frequencies have statistical significance between asthma group and control group(2=9.598,P =0.008;2=12.500,P =0.000,OR= 1.773,95%CI=1.289~2.438).compared with "CCG".allele carriers(GG、CG+GG)could elevate the risk of asthma(P = 0.012,0.002,OR= 2.469,2.406,95%CI= 1.214-5.021,1.367-4.234).Furthermore,V4 in Uygur asthma children have statistically significance(2=6.998,P =0.030),but there were no statistically significance in Han asthma children(2=4.994,P =0.082);IgE levels in asthma group were higher than those in the control group,and the difference was statistically significant(2=13.250,P =0.000);In the asthma group,IgE level in Uygur children was higher than that of Han children,the difference was statistically significant(2 =2.434,P =0.016);There were no significant differences between Han and Uygur children in control group.(2=1.072,P =0.286).No correlation were observed between ADAM33 V4 and IgE(r = 0.033,P = 0.563).Conclusion The ploymorphism of ADAM 33 V4 are related with incidence of asuima among Uygur children in Urumqi.IgE levels in Uygur children with asthma were higher than those in Han asthmatic children.Furthermore,ADAM33 V4 have not effects on IgE levels.
