摘要
目的:明确枸地氯雷他定对变应性接触性皮炎(ACD)小鼠耳廓肿胀程度的抑制作用以及对Th1/Th2相关细胞因子水平的影响。方法:建立ACD小鼠模型,观察口服不同剂量(1 mg/kg,2mg/kg,3 mg/kg,4 mg/kg)枸地氯雷他定对ACD小鼠耳廓肿胀的抑制作用(应先造模,后饲药物),采用ELISA法检测小鼠血清中IL-4、IFN-γ,TNF-α的水平。结果:口服各剂量枸地氯雷他定均能明显抑制ACD小鼠的耳廓肿胀程度,且具有剂量依赖性。模型组血清IFN-γ,IL-4,TNF-α水平明显高于正常对照组(P<0.01),药物干预后血清IFN-γ,IL-4,TNF-α水平呈剂量依赖性下降。结论:枸地氯雷他定可能通过调节Th1/Th2相关细胞因子IFN-γ,IL-4,TNF-α水平对ACD小鼠的治疗发挥作用。
Objective: To determine the effects of desloratadine citrate disodium on the edema degree of ears and the cytokine expression of Th1/Th2 cell in murine allergic contact dermatitis( ACD). Methods: The animal model of allergic contact dermatitis was established and then the inhibition effect of desloratadine citrate disodium in different doses( 1mg/kg,2mg/kg,3mg/kg and 4mg/kg) on edema degree of ACD was assessed and the expression levels of IFN-γ,TNF-α and IL-4 were detected by ELISA. Results: Desloratadine citrate disodium can reduce the edema degree of ears. The levels of IFN-γ,TNF-α and IL-4 in serum of ACD mice were higher than that in blank controls( P〈0. 01) and the levels of the three cytokines in serum were all decreased after interfered by desloratadine citrate disodium,with a dose-dependent. Conclusion:Desloratadine citrate disodium is effective in the treatment of allergic contact dermatitis,which may be through regulating the levels of IFN-γ,TNF-α and IL-4.
